摘 要:目的:观察10种中药提取物对α-葡萄糖苷酶的抑制作用。方法:以麦芽糖为底物,观察10种中药提取物(500g/L)对α-葡萄糖苷酶的抑制作用,设阿卡波糖为阳性对照。取反应液0.01mL,用氧化酶法测定生成葡萄糖的浓度;另取反应液,采用酶标仪测定吸光度,计算不同中药提取物对α-葡萄糖苷酶活性的抑制率。取3种对α-葡萄糖苷酶活性抑制作用较强的中药提取物,观察不同质量浓度下3药对α-葡萄糖苷酶的抑制率。结果:同一质量浓度下,10种中药提取物对α-葡萄糖苷酶均有抑制作用,其中青果、地榆和丹参抑制作用较强。随质量浓度(10~1000g/L)的升高,青果、地榆和丹参对α-葡萄糖苷酶活性的抑制率有升高的趋势(F=71.963、78.426和26.246,P均<0.001)。结论:青果、地榆和丹参对α-葡萄糖苷酶有较强的抑制作用。Aim:To observe the inhibitory effects of 10 kinds of traditional Chinese medicine(TCM) extracts on α-glucosidase activity.Methods:Using maltose as substrate,the inhibitory effect of 10 kinds of TCM extracts on α-glucosidase activity was observed.Acarbose was used as a positive control.The reaction solution was taken for the determination of glucose by glucose oxidase method.The absorbance of reaction solution was detected,and the inhibition rate of 10 kinds of TCM extracts on α-glucosidase activity was calculated.Three TCM extracts with higher inhibitory effect were further observed at different mass concentrations.Results:All the extracts showed inhibitory effect on α-glucosidase activity.Among them,Fructose Canarii,Radix Sanguisorbae and Radix Salviae Miltiorrhizae had higher inhibition rate.The inhibition rate of these 3 extracts increased along with the increase(10~1 000 g/L) of the mass concentration(F=71.963,78.426 and 26.246,P0.001).Conclusion:The extracts of Fructose Canarii,Radix Sanguisorbae and Radix Salviae Miltiorrhizae have relatively higher inhibitory effect on α-glucosidase activity.
