摘  要:目的:研究人抗黏液病毒A(MxA)基因重组腺病毒AdMxA体外抗乙型病毒性肝炎病毒(HBV)的作用效果。方法:以1倍感染复数(MOI)(1倍MOI感染组)及5倍MOI(5倍MOI感染组)的重组腺病毒AdMxA分别转染HepG2.2.15细胞,以未感染HepG2.2.15细胞为对照(未感染组),利用RT-PCR检测3组HepG2.2.15细胞内MxAmRNA水平表达,并检测3组HepG2.2.15细胞内HBsAg及HBeAg滴度变化。结果:3组HepG2.2.15细胞内MxAmRNA水平比较差异有统计学意义(F=226.294,P<0.001),3组细胞内HBsAg、HBeAg水平比较差异有统计学意义(F值分别为94.519和44.065,P均<0.001),而1MOI及5MOI感染组间差异无统计学意义(P>0.05)。结论:重组腺病毒AdMxA感染HepG2.2.15后其MxAmRNA表达水平明显升高,并能显著抑制HepG2.2.15细胞中HBV的复制及表达,重组腺病毒AdMxA有望成为一种有效的基因治疗方法。Aim:To investigate the anti-HBV effects with the recombinant adenovirus AdMxA in vitro.Methods:The levels of MxA mRNA were detected by RT-PCR and the tites of HBsAg and HBeAg were detected by ELASE after HepG2.2.15 cells were transfected by 1 MOI or 5 MOI of recombinant adenovirus AdMxA.And the control group was HepG2.2.15 cells without transfection.Results:The levels of MxA mRNA in HepG2.2.15 cells transfected with recombinant adenovirus AdMxA were increased significantly compared with the control group(F=226.294,P0.001).The titers of HBsAg and HBeAg in HepG2.2.15 cells transfected with recombinant adenovirus AdMxA were lower significantly than those in HepG2.2.15 cells without recombinant adenovirus AdMxA(F were 94.519 and 44.065 respectively,P0.001 in both).There was no difference between 1 MOI and 5 MOI groups in the titers of of HBsAg and HBeAg(P0.05).Conclusion:It indicates that the replication and expression of HBV in HepG2.2.15 cells can be inhibited after the recombinant adenovirus AdMxA transfected.It may be an effective way of gene therapy of HBV infection with recombinant adenovirus AdMxA.
