隔药饼灸对动脉粥样硬化兔主动脉内皮细胞过氧化酶体增殖物激活型受体γ的影响

摘　　要：目的观察隔药饼灸对动脉粥样硬化(AS)兔主动脉内皮细胞过氧化酶体增殖物激活型受体γ(PPARγ)的影响,探讨隔药饼灸延迟AS形成的作用机理。方法通过高脂饲料喂养及免疫损伤法建立兔AS模型。将75只新西兰大耳白兔随机分为5组,每组15只,即空白组(空白对照组),模型组(AS模型组),直接灸组(AS模型+艾炷直接灸),隔药饼灸组(AS模型+隔药饼灸),西药组(AS模型+阿托伐他汀)。治疗16周后检测兔主动脉内皮细胞PPARγ表达。结果模型组主动脉内皮细胞PPARγ的蛋白表达明显低于空白组、西药组和隔药饼灸组,且有统计学意义(P<0.05或P<0.01);与直接灸组比较,西药组和隔药饼灸组PPARγ平均灰度值降低(P<0.05);西药组与隔药饼灸组比较差异无统计学意义(P>0.05)。结论隔药饼灸能通过激活AS兔主动脉内皮细胞PPARγ的蛋白表达,起到延迟AS斑块形成的作用。Objective To observe the effect of herbal-cake-isolated moxibustion on the expression of peroxisome proliferator-activated receptor γ（PPARγ） in aortic endothelial cells of atherosclerosis（AS） rabbits and explore its functional mechanism in delaying the AS formation.Methods The AS models were established through high-fat diet and immunologic injury.Seventy-five New Zealand white rabbits were randomized into 5 groups,i.e.blank control group,model group,direct moxibustion group,herbal-cake-isolated moxibustion group,and atorvastatin group,with 15 rabbits in each.The expression of PPARγ in aortic endothelial cells was measured after treatment of 16 weeks.Results The PPARγ expression of the model group was lower than the blank control group,atorvastatin group,and herbal-cake moxibustion group with a significant difference（P〈0.01 or P 〈0.05）.Compared with the direct moxibustion group, the average grey degree of PPARγ of the atorvastatin group and herbal-cake-isolated moxibustion group was decreased（P〈0.05）.There was no significant difference between the atorvastatin group and herbal-cake-isolated moxibustion group（P〉0.05）.Conclusion The herbal-cake-isolated moxibustion is able to activate the PPARγ expression in aortic endothelial cells of AS rabbits to delay the formation of atherosclerotic plaque.
