摘 要:[目的]建立并应用金纳米粒子共振光散射(RLS)法,测定人尿样中金属硫蛋白(MTs)的含量。[方法]在pH4.56的B-R缓冲液中,MTs与金纳米粒子结合形成较大聚集体,导致体系RLS增强,于545nm波长处RLS强度与MTs的浓度有线性关系,据此建立测定MTs的新方法。[结果]在最佳实验条件下,在545nm波长处RLS强度与MTs在7.98×10^(-9)~1.0×10^(-7)g/mL范围内线性关系良好,其线性回归方程为△I_(RLS)=16.69p-23.09,相关系数r=0.9995,检出限为2.39×10^(-9)g/mL。[结论]本方法操作简便、快速、仪器设备简单,检出限低,灵敏度高,可用于尿中痕量MTs的测定。To establish a resonance light scattering( RLS )method for the determination of metallothioneins ( MTs ) based on the interaction of MTs with gold nanoparticles ( GNPs ). [ Methods ] In B-R buffer solution of pH 4.56, the conjugates of MTs-GNPs were formed by Au-S covalent bond between the mereapto group of eysteine of MTs and GNPs, resulting in the enhancement of RLS intensity of the system owing to their aggregating. RLS intensity linearly correlated with the concentration of MTs at 2m^x=545nm, which suggested that a new method for determination of MTs could be established. [ Results ] Under the optimum conditions, there was a linear relationship between the RLS intensity and the concentration of MTs in the range of 7.98 × 10^-9-1.0 × 10^-7 g/mL at λmax = 545 nm. The linear regression equation was ZX/RLS = 16.69,p-23.09 with a correlation eoeffieient of 0.9995, and the detection limit of 2.39 × 10^-9g/mL. [ Conclusion ] This method is simple, rapid, sensitive and accurate, and the detection limit is also low. It has been applied for deteeting traee MTs in urine.
