摘 要:目的:探讨赤丹丸中其他成分对君药赤芍中芍药苷在大鼠体内药动学过程的影响。方法:分为赤芍组、赤丹丸组,分别灌胃给予赤芍煎煮液、赤丹丸煎煮液,设定时间点采血,沉淀蛋白法提取血浆中的芍药苷,以栀子苷为内标,采用HPLC法测定大鼠血浆中芍药苷的质量浓度,DAS数据处理软件计算药动学参数。结果:给予赤丹丸组与单独给予赤芍组相比,芍药苷在大鼠体内tmax,CL,MRT0→τ降低,t1/2,Cmax,V,MRT0→∞,AUC0→τ和AUC0→∞均升高,其中tmax,Cmax和t1/2的变化具有显著性差异(P<0.05,P<0.01)。结论:赤丹丸中其他成分可显著影响君药赤芍中主要成分芍药苷的药动学。可为中药复方制剂在体内作用机制的研究提供思路,并指导临床用药情况。OBJECTIVE To study the effects of other ingredients in Chidan pills on paeoniflorin of paeoniflorin (Radix Paeoniae Rubra) with the plasma of rats. METHODS Decoctions of Radix Paeoniae Rubra and Chidan pills were orally administered to SD rats respectively. Plasma samples collected at selected time points were pretreated by protein precipitation. And geniposide was used as the internal standard. High performance liquid chromatography was applied to determine the concentration of paeoniflorin in rat plasma. The data were processed by DAS to calculate the pharmacokinetie parameters. RESULTS The parameters of Tmax,CL and MRT0→τ were decreased when orally administered the decoctions of Chidan pills, but t1/2, Cmax,V, MRT0→∞, AUC0→∞ and AUC0→∞ were increased with comparison of the decoction of Radix Paeoniae Ruhra. There were statistically significant differences in pharmacokinetic parameters of paeoniflorin including the Tmax ,Cmax and t1/2 (P〈0. 05, P〈0. 01). CONCLUSION Other ingredients in Chidan pills could influence the pharmacokinetic process of paeoniflorin in Radix Paeoniae Rubra. This study can provide the thoughts for the mechanism of the Chinese traditional medicine compound preparation in vivo and guide the clinical use of medicines.
