摘 要:目的:探讨美他多辛对雄性酒精性脂肪肝大鼠肝组织中CYP2E1表达的影响。方法:30只清洁级Wistar大鼠随机分为3组:脂肪肝组梯度给予不同浓度的酒精灌胃8周;预防组于酒精灌胃同期给予美他多辛预防;对照组给予普通饮食。8周酒精灌胃结束后,B超检测肝组织影像学变化、病理学观察酒精性脂肪肝的形成以及免疫组织化学检测肝组织中CYP2E1的表达。结果:雄性酒精性脂肪肝大鼠模型建立成功;脂肪肝组、预防组与对照组比,CYP2E1表达均上调,P<0.05;脂肪肝组与预防组比,CYP2E1表达上调,P<0.05。结论:CYP2E1表达的上调参与了酒精性脂肪肝的发病过程,美他多辛可有效预防酒精性脂肪肝的发生及CYP2E1表达的上调。OBJECTIVE To investigate the effects of metadoxine on the expression of CYP2E1 in liver tissue of male rats with alcoholic fat liver. METHODS 30 clean-rank Wistar rats were randomly divided into three groups: alcoholic fat liver group,given different concentrations of ethanol for 8w; the prevention group, fed alcohol and given metadoxine: the control group,given normal diet. At the end of 8w, all the rats were check with the B-test, the pathological changes were observed in the formation of alcoholic liver, and the expression of CYP2E1 in liver tissue were observed by the immunohistochemical detection. RESULTS The model of male rats with alcoholic fat liver was set up successfully; in alcoholic fat liver group and the prevention group the expression of CYP2E1 was higher than that in the control group, P〈0. 05;as compared with alcohol fat liver group,in the prevention group, the expression of CYP2E1 was decreased, P〈0. 05. CONCLUSION Up-regulation of CYP2E1 expression involved in the pathogenesis of alcoholic fat liver disease, metadoxine plays an effective role in preventing the occurrence of alcoholic fat liver disease, and influences on the expression of CYP2E1.
