大孔吸附树脂纯化黄精小分子糖的工艺研究

摘  要:目的:研究大孔树脂吸附纯化黄精小分子糖的最佳工艺条件及参数。方法:以小分子糖保留率和纯度为考察指标,比较四种大孔树脂对黄精小分子糖的吸附性能,考察上样浓度、上样量、洗脱流速和洗脱剂用量等因素对优选树脂纯化工艺的影响,确定最佳工艺参数。结果:最佳工艺条件为选用AB-8大孔吸附树脂,上样浓度36.32 mg·ml^(-1),最大上样量4倍柱床体积,以蒸馏水8倍柱床体积洗脱,洗脱流速为1.0 ml·min^(-1)。通过本工艺纯化的黄精小分子糖保留率达97.56%,纯度达90.93%。结论:大孔吸附树脂对黄精小分子糖有较好的纯化作用,该工艺简单可行,适合于工业化生产。Objective : To study the optimal conditions and parameters of purifying low molecular weight saecharide in Rh/zoma polygonati with macroporous resins. Method : With the yield and purity as the indices, the absorption characteristics of four resins for low molecular weight saccharide purification were compared. The optimal conditions of the purification technology were investigated. Result: The AB-8 resin was the best of the four resins and the optimum conditions were the following: solution concentration of 36. 32 mg'm1-1, sample volume of 4 times of resin volume, eluting cubage of 8 times of resin volume, distilled water as eluting solvent with eluting rate of 1.0 ml·min-1. The yield was 97. 56% ,and the purity reached 90. 93%. Conclusion: The process of macroporous absorption resins to purify low molecular weight saccharide is simple, feasible and suitable for industrial production.
