HPLC法测定茵杖舒胆颗粒与合剂中4种大黄蒽醌的含量

摘　　要：目的:建立茵杖舒胆颗粒剂与合剂中大黄酸、大黄素、大黄酚和大黄素甲醚的HPLC含量测定方法。方法:采用VenusilXBP-C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相:甲醇-0.1%磷酸溶液(85:15),检测波长:254 nm,流速:1.0 ml.min^(-1),柱温:30℃。结果:4种大黄蒽醌均呈良好的线性(r=0.999 9～1.000 0),茵杖舒胆颗粒的平均回收率为96.5%～100.3%(RSD=2.18%～3.07%);茵杖舒胆合剂的平均回收率为97.1%～100.3%(RSD=1.69%～3.00%)。结论:通过对茵杖舒胆颗粒和合剂各6批的含量测定,证明方法简单,准确可靠,重复性好。Objective： To develop a quantitative reversed phase high performance liquid chromatography method for simultaneous determination of Rhein, Emodin, Chrysophanol and Physcion in Yinzhang Shudan granules and mixture. Method ： The samples were separated at 30℃ on a Venusil XBP-C18 （250 mm × 4. 6 mm ,5 μm） column eluted with methanol-0. 1% phosphate solution （85：15 ） as the mobile phase. Flow rate was set at 1.0 ml. min-1 and the detection wavelength was set at 254 nm. Result： The linear ranges of the anthraquinones were wide ,showing good linear correlations with correlation coefficients （r） of 0. 999 9 to 1. 000 0. The average re- coveries of granules were from 96. 5 % to 100. 3% （RSD =2. 18%-3.07% ） ,and the average recoveries of mixture were from 97.1% to 100. 3% （RSD = 1.69%-3.00% ）. Conclusion： The developed method was proved to be simple,reliable and useful in analysis of 6 batches of the granules and mixture samples, respectively.
