摘 要:目的:探讨桦褐孔菌乙醇提取物对哮喘小鼠气道炎症和Thl/Th2类细胞因子变化的影响及其可能机制。方法:雄性BALB/c小鼠32只随机分成4组,即正常对照组、哮喘模型组和桦褐孔菌乙醇提取物低、高剂量干预组。分别采用酶联免疫吸附法(ELISA)和蛋白质印迹检测支气管肺泡灌洗液(BALF)中IL-4,IL-5,IL-13,IFN-γ含量以及肺组织中磷酸化p38MAPK的变化,并观察BALF中炎症细胞和肺组织病理学改变。结果:哮喘模型组小鼠BALF中炎症细胞计数,IL-4,IL-5,IL-13水平和IFN-γ水平降低;肺组织中磷酸化p38 MAPK表达水平升高,与正常组比较差异显著(P<0.05)。桦褐孔菌乙醇提取物低、高剂量干预组小鼠BALF中炎症细胞计数,IL-4,IL-5,IL-13水平和肺组织中磷酸化p38MAPK表达水平均明显降低,IFN-γ水平明显上升,与哮喘模型组比较均具有显著差异(P<0.05)。桦褐孔菌乙醇提取物处理能明显减轻哮喘小鼠肺组织病理学改变,桦褐孔菌低、高剂量干预组之间比较无显著差异。结论:p38 MAPK可能参与了支气管哮喘的发病过程。桦褐孔菌对哮喘小鼠具有保护作用,其机制可能与抑制p38 MAPK磷酸化、调节IL-4/IFN-γ平衡失调、减轻炎症细胞浸润有关。Objective: To explore the protective effects and mechanism of ethanol extract of Inonotus obliquus (EEIO) injection on asthmatic mice. Method: OVA was injected intraperitoneally and inhaled to produce the asthmatic model. Thirty two mice were randomly divided into four groups : control group, asthma group and 1. obliquus groups of high and low dose. The concentrations of IL- 4,IL-5 ,IL-13 and IFN-γ in BALF, the phosphor-p38 MAPK in lung tissues were respectively measured by ELISA and Western blotting. The number of inflammatory cells in BALF and histopathology changes were observed. Result: In asthmatic group,the number of inflammatory cells and the concentrations of IL-4, IL-5, IL-13 in BALF and phospho-p38 MAPK in lung tissue were higher, while IFN-γ were lower than those in normal control mice ( P 〈 0. 05 ). In 1. obliquus group, the number of inflammatory cells, the concentrations of IL-4,IL-5 ,IL-13 in BALF and phosphor-p38 MAPK in lung tissue were lower, but were higher than those in normal control mice ( P 〈 0.05 ), and histropathology damage was alleviated significantly. There was no significant difference observed among the efficacies in the I. obliquus groups of high and low dose. Conclusion: p38 MAPK may play a role in pathological process of asthma. I. obliquus effectively treats asthma by inhibiting the expression of phosphor-p38 MAPK, correcting the unbalance of IFN-3,/IL-γ and decreasing the number of inflammatory cells.
