摘 要:目的:观察姜黄素对APPswe/PS1dE9双转基因小鼠Aβ42及其降解酶NEP表达的影响。方法:将3月龄的APP-swe/PS1dE9双转基因小鼠随机分为模型组、阳性对照组、姜黄素高、中、低剂量组。灌胃3个月后,应用Morris水迷宫、免疫组织化学、Western blot等方法进行检测。结果:行为学检测,治疗组与模型组相比定位航行实验和空间探索实验存在不同程度的差异(P<0.01或P<0.05)。Aβ42及其降解酶NEP表达,模型组小鼠大脑海马CA1区较正常对照组Aβ42阳性细胞明显增加(P<0.01)。结论:姜黄素能通过增加Aβ42降解酶NEP表达、降低Aβ42蛋白,进而改善APPswe/PS1dE9双转基因小鼠的学习记忆能力。Objective: To observe the effect of curcumin on the expression of Aβ42 and its degrading enzyme NEP in APP/PS1 double transgenic mice. Method: 3-month old APP/PS1 dig mice were randomly divided into model group, positive control group and curcumin large, medium and small dose group. After 3 months, Morris water maze, immunohistochemistry, Western blot were applied to detect learning and memory ability of animal. Result: Behavior detection, compared with the model group, treatment group showed varying degrees of difference in place navigation test and space exploration experiments( P 〈 0. 01 or P 〈 0. 05 ). The expression of Aβ42 and its degrading enzyme NEP, Aβ42-positive cells of hippocampus CAI region significantly increased in model mice as compared with normal control group(P 〈 0.01 ). Conclusion: Curcumin can improve learning and memory ability of APP/PS1 double transgenic mice through increasing the expression of Aβ-degrading enzyme NEP and decreasing the expression of Aβ42.
