摘 要:目的:探讨人羊膜上皮细胞(humanamniotic epithelial cells,hAECs)体外向神经样细胞诱导分化的条件及可能性。方法:利用碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)、化学诱导剂(alltransretinoi cacid,RA)诱导人羊膜上皮细胞(WISH)向神经元转化,并以不含诱导剂的无血清的MEM培养基作为对照,观察分化过程中细胞形态的变化,利用免疫细胞化学方法检测神经元特异性标志物的表达情况。结果:诱导分化12h后的细胞呈现双极、多极和锥形的典型神经元细胞的形态,免疫细胞化学显示诱导后的hAECs表现为NSE染色阳性,不表达神经胶质细胞标记物GFAP。结论:hAECs可以在体外诱导分化为神经元样细胞,这种潜能使其有可能成为神经系统疾病细胞移植治疗的种子细胞。Objective To explroe the possibility and conditions of in vitro differentiation of human amniotic epithelial cells (hAECs) into neuron-like cells. Methods Basic fibroblast growth factor and alltransretinoic acid, a chemoattractant, were used to induce differentiation of hAECs, and MEM without inducer and fetal bovine serum was used as control. The change in cell morphology was observed, and the expressions of the specific markers of neurons were detected by immunohistochemistry. Results After inducement, hAECs turned into neurons-like cells with typical bipolar, multipolar and cone-shaped manifestation, being positive with NSE staining but not expressing GFAP, a marker of glial cells. Conclusions hAECs can be induced to differentiate into neuron-like cells in vitro, whose potential may becomes a novel candidate resource for cellular transplant therapy of nervous system diseases.
