摘  要:目的分析新疆乌鲁木齐市2008年流行的11株成人麻疹野病毒的基因特征。方法用Vero/slam细胞从成人麻疹暴发患者的标本中分离出麻疹野病毒,应用逆转录一聚合酶链反应(RT-PCR)对麻疹野病毒分离株扩增核蛋白N基因羧基末端676个核苷酸片段,再对扩增产物进行核苷酸序列测定和分析。并以N基因羧基末端450个核苷酸片段构建基因亲缘性关系树,进行核苷酸变异分析。结果共分离到1l株麻疹野病毒,这些病毒N基因450个核苷酸之间的差异为0%~0.2%,均为H1基因型中的Hla基因亚型。11株麻疹野病毒与H1基因型代表株(Chin9372)的核苷酸和氨基酸差异分别为2.2%~2.4%之间和4.0%~4.6%之间。结论 2008年新疆乌鲁木齐流行的成人麻疹野病毒为Hla基因亚型。Aim To analyze the genetic characteristics of 11 measles virus strains prevalent in Urumuqi in 2008.Methods Measles viruses were isolated from adult measles cases with Vero/slam cells and the 676 nucleotide fragments at nucleoprotein N gene carboxyl terminal were amplified by reverse transcription polymerase chain reactions(RT-PCR),then the amplified sequences were identified and analyzed.The genetic sibling phylogenetic tree was constructed wih 450 nucleotide fragments at N-gene carboxyl terminal and the variation of nucleotide was analyzed.Results Totally 11 measles virus strains were isolated and the differences of among 450 nucleotide of virus N-gene were 0.%~0.2%,all belonged to Hla-gene subtype.The differences of nucleotide and amino acid of 11 measles virus strains and the representative H1 genotype strains(Chin9372)were between 2.2%~2.4% and 4.0%~4.6%.Conclusion The adult wild measles virus strains prevalent in Urumuqi in 2008 was Hla genetic subtype.
