摘  要:目的制备和鉴定B型流感病毒单克隆抗体(McAb),建立斑点金免疫渗滤法(DIGFA),用于B型流感病毒的临床检测。方法以B型流感病毒抗原免疫BALB/c小鼠制备McAb,利用胶体金标记技术对抗体进行标记,建立快速检测B型流感病毒抗原的斑点金免疫渗滤法,并对实验条件进行探讨。用建立的DIGFA法与酶联免疫吸附试验(ELISA)同时检测128例患者鼻咽部分泌物,比较敏感性、特异性,以评价检测效果。结果 DIGFA法与ELISA法对比检测了128例样本中B型流感病毒抗原,本方法的敏感性为94.4%,特异性为96.4%,两法总符合率为96.1%。结论 DIGFA法操作简单、方便、快速、特异、灵敏,在B型流感病毒感染的辅助诊断中具有较好的应用价值。Aim To prepare and characterize monoclonal antibodies(McAbs) against influenza B virus,establish the dot immunogold filtration assay(DIGFA) for detecting influenza B virus.Methods BALB/c mice were immunized with influenza B virus antigen for producing McAbs.With the colloidal gold marked McAbs,a rapid DIGFA-based method was established and the optimal experimental conditions were discussed.A total of 128 secretion samples from nasopharyngeal patients were detected by enzyme-linked immunosorbent determination method(ELISA) and DIGFA.The specificity and sensitivity of the two methods was compared.Results A total of 128 patients with influenza B virus were detected by DIGFA and ELISA.The specificity and sensitivity of DIGFA were 94.4% and 96.4%.The coincidence rate of two tests was 96.1%.Conclusion DIGFA is a simple,convenient,rapid,specific and sensitive method for detecting influenza B virus.
