摘  要:目的建立罗勒中芦丁和槲皮素的含量测定方法,并对不同产地药材进行含量测定。方法采用高效液相色谱法将罗勒中芦丁和槲皮素进行测定;芦丁的流动相为乙腈-0.4%磷酸(18∶82),槲皮素的流动相为乙腈-0.4%磷酸(32∶68);检测波长均为360 nm;流速均为1.0 mL·min-1;柱温均为30℃。结果芦丁的线性范围为4.95~24.75μg(r=0.999 7),平均回收率为100.7%,RSD=1.62%(n=6)。槲皮素的线性范围为1.792~8.064μg(r=0.999 7),平均回收率为98.3%,RSD为0.94%(n=6)。不同产地罗勒药材中芦丁和槲皮素的含量分别在0.39~0.71 mg·g-1和0.29~0.58 mg·g-1之间。结论本法专属性强,重复性好,结果准确可靠,可作为罗勒的质量控制方法。OBJECTIVE To establish a method for determination of the contents of rutin and quercetin in Ocimi Herba,and to determine the contents of rutin and quercetin in Ocimi Herba from different places.METHODS The contents of rutin and quercetin in Ocimi Herba were determined by HPLC.The rutin was determined by using mobile phase consisted of acetonitrile-0.4% phosphoric acid solvent(18∶82),the quercetin was determined by using mobile phase consisted of acetonitrile-0.4% phosphoric acid solvent(32∶68).All the UV wavelength was 360 nm,the flow rate was 1.0 mL·min.1 and the column temperature was 30 ℃.RESULTS The linear range of rutin was 4.952-4.750 μg(r=0.999 7) with a mean recovery of 100.7% and RSD 1.62%(n=6).The linear range of quercetin was 1.792.8.064 μg(r=0.999 7) with a mean recovery of 98.3%,RSD 0.94%(n=6).The contents of rutin and quercetin in Ocimi Herba from different places were within the range of 0.39.0.71 mg·g.1 and 0.29.0.58 mg·g.1.CONCLUSION The method is specific,reliable and accurate.It can be used for the quality control of Ocimi Herba effectively.
