黄芪注射液对两种急性心衰模型的强心作用比较

摘　　要：目的:观察比较黄芪注射液和经典洋地黄类强心药去乙酰毛花苷注射液对戊巴比妥钠和心得安所致不同大鼠急性心衰模型的强心作用。方法:48只健康Wistar大鼠随机分为戊巴比妥钠模型组、戊巴比妥钠模型去乙酰毛花苷组、戊巴比妥钠模型黄芪注射液组、心得安模型组、心得安模型去乙酰毛花苷组和心得安模型黄芪注射液组,每组8只。分别于静脉输入戊巴比妥钠或心得安制作大鼠急性心衰模型,戊巴比妥钠模型去乙酰毛花苷组和心得安模型去乙酰毛花苷组在造模成功后均给予去乙酰毛花苷注射液(0.1 mg.kg-1),戊巴比妥钠模型黄芪注射液组和心得安模型黄芪注射液组在造模成功后均给予黄芪注射液(4 ml.kg-1),检测各组大鼠在造模成功并稳定20min后,静脉注射去乙酰毛花苷注射液或黄芪注射液5min、15min、30min、60min和120min各时间点LVSP、+dp/dtmax、HR等左室收缩功能的变化。结果:2种造模方法均能使大鼠LVSP、+dp/dtmax、HR显著降低,且在2h内保持在一个较低水平,显示大鼠急性心衰模型制作成功而稳定;静脉注射黄芪注射液使2种模型大鼠的LVSP、+dp/dtmax均显著升高,心得安模型大鼠黄芪注射液起效更为迅速,于药后5min即出现了明显的正性肌力作用(P<0.05);戊巴比妥钠模型大鼠药后15min才出现心肌收缩力的明显增强(P<0.05)。结论:与戊巴比妥钠所致急性心衰模型相比,心得安诱发的急性心衰模型更适合于黄芪注射液治疗急性心衰的药理药效学研究。Objective：To compare the cardiotonic effect of astragalus injection in the two acute heart failure（HF） models induced by pentobarbital sodium and propranolol respectively,so as to determine the suitability of the models for the study of astragalus injection.Method： The acute HF models were created by pentobarbital sodium and propranolol respectively in vivo.Astragalus injection was then injected intravenously into the rats with the two different models.LVSP、 ＋ dp /dtmax and HR were recorded throughout the experiment.Results： The two kinds of acute heart failure models were created successfully.LVSP and ＋ dp /dtmax in two models were increased after astragalus injection was injected intravenously.After injection of astragalus injection,both LVSP and ＋ dp /dtmax were increased more rapidly in the propranolol-induced HF model as compared with that in pentobartital sodium-induced HF model.Conclusion：Propranolol-induced acute HF is comparatively more suitable for the pharmacological study on astragalus injection.
