电针对局灶性脑缺血模型大鼠脑内STAT5阳性神经元的影响

摘　　要：目的:了解STAT5在局灶性脑缺血后的变化及电针干预对其的影响。方法:采用热凝法制成MCAO模型,采用免疫组织化学的方法检测不同时间段STAT5在病灶侧皮质、海马CA1区的水平。结果:造模2h后各指标明显升高,在多个指标上与假手术组相比有显著性差异(P<0.05);24h后数值降低,3d后降低接近假手术水平。电针组的检测结果与模型组的趋势相同,但与模型组相较,各时间段的数据均高于相应的模型组。结论:电针治疗在脑缺血超早期就能明显提高STAT5水平,激发机体的自我保护机制,同时还可以激发这种保护机制持续作用达3d以上。Objective： To detect STAT5-immunoreactive cell levels in the cortex and hippocampus CA1 surrounding the cerebral ischemia area in model rat brain and effects of electro-acupuncture.Methods： Model of MCAO was made with thermocoagulation and level of STAT5-positive neurons was detected in the cortex and hippocampus CA1 area at various time periods by using immunohistochemical method;Results： Compared to sham-operated group,the various indicators increased significantly after making model 2 hours and there was significant difference on a number of indicators（P 0.05）.Their data decreased after 24 hours and be to further reduce after 3 days,which close to the level of sham-operated.The results of electro-acupuncture group have the same trend with model group,namely,the highest was 2-hour group,followed by a 24-hour group,3-day group descending further.However,the data of each group were higher than the corresponding model group,there are clear advantages.Conclusion： The electro-acupuncture treatment can markedly improve ultra-early STAT5 levels after cerebral-ischemia,stimulate the body’s self-protection mechanisms,can also stimulate role of such a protective mechanism to continue to 3 days or more.
