摘  要:目的研究CCK-8法检测兔筋膜成纤维细胞(RFFBs)增殖活性的最佳测试条件。方法以兔筋膜作为成纤维细胞原代培养组织,利用传代至P3的成纤维细胞,对比CCK-8法在不同孵育时间及不同检测波长条件下吸光光度值的差异,研究CCK-8法检测兔筋膜成纤维细胞增殖活性的最佳检测时间及最佳检测波长。结果 CCK-8法检测兔筋膜成纤维细胞增殖活性的最佳检测时间为加入CCK-8试剂孵育后的4 h,最佳检测波长为450 nm。结论 CCK-8法在孵育4 h后及酶标仪450 nm波长下检测兔筋膜成纤维细胞的灵敏度最高。Objective To research the best test conditions of CCK-8 method detecting rabbit fascia fibroblasts'(RFFBs) proliferation activity.Methods With rabbit fascia tissue as fibroblasts former generation training organization,using the fibroblasts of P3 generation,comparing the difference of OD value between different kinds of test times or different kinds of test wavelengths.Researching the best test time and test wavelength with CCK-8 method detecting rabbit fascia fibroblasts'(RFFBs) proliferation activity.Results Using CCK-8 method to detect rabbit fascia fibroblasts'(RFFBs) proliferation activity,the best test time is 4 h after adding CCK-8 reagent,the best test wavelength is 450 nm.Conclusion The sensitivity is highest with CCK-8 method in 4 h after adding CCK-8 and 450 nm wavelengths detecting rabbit fascia fibroblasts.
