促红细胞生成素对阿霉素诱导的心肌细胞凋亡的预防作用研究

摘　　要：目的:研究促红细胞生成素(Erythropoietin,EPO)预防阿霉素(Doxorubicin,DOX)诱导的心肌细胞凋亡的作用。方法:原代培养乳鼠心肌细胞,随机分为空白对照组、模型对照组及不同浓度(6.25、25、100 IU/ml)EPO预处理组,各组预处理24 h后加入5μg/ml的DOX。24 h后采用流式细胞术检测细胞凋亡率,甲基噻唑基四唑(Methyl thiazolyl tetrazolium,MTT)法检测细胞活力,流式细胞术检测凋亡蛋白Bcl-2和Bax的表达。结果:与空白对照组相比模型对照组细胞活力明显减低,凋亡率显著增高。不同浓度EPO预处理的心肌细胞活力较模型对照组显著增高,细胞凋亡率显著降低,并且凋亡抑制蛋白Bcl-2的表达增高,凋亡蛋白Bax的表达降低。结论:EPO对DOX诱导的心肌细胞的损伤具有预防作用,其机制可能与调节凋亡相关蛋白Bcl-2和Bax有关。Objective：To investigate the preventive effect of erythropoietin（EPO） on doxorubicin（DOX）-induced apoptosis in myocardial cells of rats.Methods：Cells were cultured in vitro and treated with different concentrations（6.25,50,100 IU /ml） of EPO for 24 hours,then the myocardial cells were exposed to DOX（5 μg/ml）.After 24 hours,the apoptosis ratio was detected by flow cytometry,and cell viability was detected by methyl thiazolyl tetrazolium.The expression of apoptotic protein Bcl-2/Bax was detected by flow cytometry.Results：The antiapoptotic effect of EPO was in a concentration manner.The survival rate of cells was increased,and the apoptosis ratio decreased.Conclusion：EPO has protective effect against DOX-induced apoptosis in myocardial cells.EPO may increase the expression of apoptosis protein Bcl-2/Bax and decrease the apoptosis ratio.
