摘 要:目的:用酒精联合内毒素建立一个新型的慢性胰腺炎(Chronic pancreatitis,CP)的大鼠模型,为研究CP的发病机制提供合适的动物模型。方法:18只1月龄雄性SD大鼠随机分为对照组、酒精组和模型组,每组各6只。饲以25%酒精,12周后反复腹腔注射脂多糖(Lipopolysaccharides,LPS),建立CP的动物模型。在光镜下观察胰腺组织显微结构变化,用Masson染色法观察胰腺组织胶原的改变,并测定胰腺组织匀浆中的淀粉酶和脂肪酶及血糖浓度。结果:模型组胰腺小叶内可见炎症细胞浸润,胶原纤维较对照组和酒精组明显增多,胰腺组织内淀粉酶和脂肪酶降低(P<0.05)。结论:在长期摄入乙醇的基础上反复腹腔注射LPS可造成CP发生,本模型可以用于CP的研究。Objective:To investigate the pathogenesis of chronic pancreatitis(CP) by establishing a new pancreatitis model in rats suitable for investigation of the processes and mechanisms leading to pancreatic fibrosis.The model was established with ethanol and lipopolysaccharides(LPS,endotoxin).Methods:Eighteen one-month old male SD rats were randomly divided into control group,alcohol group,and model group.After fed with 25% alcohol for twelve weeks,the rats in model group were injected with lipopolysaccharides(LPS).Pancreatic morphology was conducted with haematoxylin-eosin stain,collagen deposition was determined by Masson stain,and the amylase and lipase in pancreatic tissue homogenate were evaluated by colorimetry.Results:Compared with control group,the acinar cells with vacuolization,inflammatory cell infiltration and collagen increasing were observed in the model group.Lipase and amylase were significantly decreased in the model group(P0.05).Conclusion:Combined with ethanol,LPS can induce chronic pancreatitis.The rat model may be useful for studying the mechanism of chronic pancreatitis.
