摘 要:以MS、LS、B5、N61、/2 MS、Nitsch、WPM和White为基本培养基,添加0.2 mg·L^-1NAA+0.5 mg·L^-16—BA,分析不同类型培养基对华中五味子愈伤组织生长及其多糖含量的影响,并以MS培养基进行生长调节剂水平、碳源、蔗糖质量浓度及光照条件试验,探讨不同培养条件的影响。结果表明:MS、LS、1/2 MS培养基适合愈伤组织生长,其中1/2 MS培养基有利于多糖的积累;WPM和White培养基既不适合愈伤组织生长也不利于多糖的积累;较低质量浓度组合的0.2-0.5 mg·L^-16—BA+0.2-0.5 mg·L^-1NAA有利于愈伤组织的生长,而较高质量浓度组合的1.0-2.0 mg·L^-16—BA+1.0-2.0 mg·L^-1NAA则有利于多糖的积累;作为培养基的碳源,蔗糖比葡萄糖更有利于愈伤组织的生长,而葡萄糖比蔗糖更有利于多糖的积累;培养基中蔗糖质量浓度对愈伤组织生长有明显影响,对多糖积累的影响不大,合适的蔗糖质量浓度为25 g·L^-1;光照培养与暗培养对愈伤组织增殖及多糖含量的影响均无明显差异。The MS,LS,B5,N6,1/2 MS,Nitsch,WPM and White,adding with 0.2mg·L^-1 NAA + 0.5mg·L^-1 6-BA respectively,were used as basic media to study their effects on callus growth and polysaccharide production in Schisandra sphenanthera.Growth regulators,carbon sources,sucrose concentration and light were also studied with MS as basic medium.The results show that MS,LS and 1/2 MS media were suitable for callus growth and 1/2 MS medium was conducive to polysaccharide accumulation;WPM and White media were neither suitable for callus growth nor conducive to polysaccharide accumulation.It was indicated that growth regulator combination of 0.2~0.5mg·L^-1 6-BA + 0.2~0.5mg·L^-1 NAA was suitable for callus growth,while the combination of 1.0~2.0mg· L^-1 6-BA+1.0~2.0mg·L^-1 NAA was conducive to accumulation of polysaccharide;as carbon source of a medium,sucrose was more conducive to callus growth than glucose,whereas glucose was more favorable to polysaccharide accumulation than sucrose;sucrose concentration had a significant effect on the accumulation of polysaccharide but little effect on callus growth;the appropriate sucrose concentration was 25g·L^-1;the callus growth and polysaccharide content were of no significant difference between light culture and dark culture.
