摘  要:目的观察虎杖苷(PD)对颈动脉窦压力感受器反射的影响,并探讨其可能的作用机制。方法将大鼠随机分为正常组、PD 50μmol/L组、PD 100μmol/L组和PD 200μmol/L组。应用隔离灌流麻醉大鼠颈动脉窦区的技术,观察不同浓度的PD(50,100,200μmol/L)对颈动脉窦压力感受器反射(CSB)的影响。结果与正常组大鼠相比,PD组大鼠压力感受器曲线明显向右上方移位,且具有剂量依赖性。压力感受器反射曲线最大斜率(PS)和MAP反射性下降的最大值下降。Bay K8644(500 nmol/L)预处理后,PD抑制CSB的效应被完全阻断。结论 PD可抑制CSB,并且此效应与阻断钙通道有关。Objective It is to study the effect of Polydatin(PD) on carotid sinus baroreflex(CSB) and explore the underlying mechanism.Methods The rats were randomly divided into normal group,PD 50 μmol/L group,PD 100 μmol/L group and PD 200 μmol/L group.The effect of PD with different content(50,100,200 μmol/L) on CSB was observed by perfusing the isolated carotid sinus of the rats.Results Compared with that in normal group,PD shifted the functional curve of CSB to the right and downward,with a marked decrease in peak slope(PS) and reflex decrease(RD) in a concentration-dependent manner,indicating the inhibitory effect of PD on CSB.Pretreatment with Bay K8644(500 nmol/L) could completely abolish the effect of PD on CSB.Conclusion PD can inhibits CSB through blocking Ca2+ channels in carotid sinus of anesthetic rats.
