摘 要:目的:抗双链DNA(dsDNA)抗体的诱生机理不明,试图研究诱导该自身抗体产生的相应自身抗原。方法:取体外活化的Balb/c小鼠脾细胞及其染色质免疫同系小鼠,用ELISA方法检测IgG类抗双链DNA抗体及其IgG亚类。结果:不同原因活化的脾细胞及其染色质均可诱导IgG类抗双链DNA抗体产生,且抗体均为IgG1亚类。结论:活化淋巴细胞及其活性染色质是诱导双链DNA抗体生成的自身免疫原。Objective: The relevant primary autoantigen which induced antidsDNA antibodies was still enigmatic.Attempted to investigate the primary autoantigens driving the productiorn of these autoantibodies. Methods: Immunized Balb/c mice with activated syngenic spleen cells primed by ConA or LPS or rIL2 in vitro and the chromatin isolated from activated spleen cells.AntidsDNA IgG antibodies and its subclass were determined by ELISA. Results: The activated lymphocytes and its chromatin all induced antidsDNA antibodies and these autoantibodies belong to IgG1 subclass. Conclusion: The results indicated that activated lymphocytes and their active chromatin can be the autoimmunogenes driving the production of antidsDNA antibodies.
