摘 要:目的 观察一氧化氮 (NO)在白细胞介素 (IL) 1β、IL 6对胰岛细胞影响中的作用。 方法 应用体外单层培养的大鼠胰岛细胞检测IL 1β、IL 6对胰岛细胞亚硝酸盐生成、胰岛细胞内DNA、胰岛素含量和细胞活性的影响 ,并进一步观察一氧化氮合酶抑制剂NG 单甲基 L 精氨酸 (L NMMA)的作用。结果 以IL 1β诱导 ,胰岛细胞亚硝酸盐生成量显著增加 (P <0 .0 0 1) ;同时胰岛细胞内DNA、胰岛素含量及细胞活性 (MTT值 )均显著下降 (P <0 .0 0 1) ;L NMMA有阻抑作用 (P <0 .0 0 1)。IL 6不能诱导胰岛细胞的NO生成 ,亦不影响细胞内DNA、胰岛素含量及细胞活性 (P >0 .0 5 )。IL 1β与IL 6联合作用时 ,胰岛细胞亚硝酸盐生成量显著增加 ;胞内DNA、胰岛素含量及MTT值均显著下降 (P <0 0 0 1) ,但与IL 1β单独作用差异无显著性 (P >0 .0 5 )。 结论 IL 1β通过NO介导 ,对胰岛细胞有细胞抑制和细胞毒性效应。IL 6对胰岛细胞无直接的毒性作用 ,这可能与其不能诱导NO的生成有关。Objective To observe the effects of interleukin (IL) 1β and IL 6 on nitric oxide (NO) production, islet cell DNA and insulin content, and MTT assay of rat pancreatic islets of Langerhans. Methods Isolated pancreatic islet cells from SD rats were cultured in monolayer in vitro. Nitrite production, islet cell DNA and insulin content, and MTT assay in rat pancreatic islet cells incubated with IL 1β and IL 6, singly or in combination, were measured. The levels of these parameters of the cytokines in the absence and presence of N G monomethyl L arginine (L NMMA), an inhibitor of nitric oxide synthetase, were compared. Results IL 1β induced a significant increase in nitrite production, a decrease of islet cell DNA and insulin content and reduction of cell activity (MTT, P <0.001); these changes were blocked by L NMMA ( P <0.001). IL 6 produced no significant change in those parameters ( P >0.05). The combination of IL 1β and IL 6 increased NO production, decreased islet cell DNA and insulin content and reduced cell activity ( P <0.001); the changes were similar to those with IL 1β alone ( P >0.05). Conclusion The cytotoxic effects of IL 1β on pancreatic islets might be mediated by NO. IL 6 may not be directly cytotoxic to pancreatic islets. It may be related with the fact that IL 6 does not induce NO production.
