摘 要:目的观察丝瓜络(RLF)对慢性充血性心衰大鼠尿量及肾脏水通道蛋白-2(AQP-2)基因表达的影响,初步探讨RLF的利尿作用及其分子生物学机制。方法选取Wista大鼠,采用异丙肾上腺素法建立慢性充血性心衰大鼠模型,分别给予RLF5g·kg-1·d-1,10g·kg-1·d-1,灌胃给药,1次/d,连续用药28d。采用代谢笼法,测定各组大鼠24h尿量;采用实时荧光定量逆转录聚合酶链反应(RT—PCR)技术,观察各组大鼠肾脏髓质AQP-2mRNA的表达。结果丝瓜络连续用药可显著增加心衰大鼠尿量并降低心衰大鼠肾脏髓质AQP-2mRNA的表达(P〈0.05)。结论丝瓜络对慢性充血性·心衰大鼠有明显利尿作用,其分子生物学机制可能与降低肾脏髓质AQP-2mRNA的表达有关。Objective To observe the effects of Retinerus Luffae Fruetus(RLF) on the urine volume and the renal medullary Aquaporin-2(AQP-2) mRNA expression in rats with chronic congestive heart failure,and to explore the diuretic effect and its molecular mechanism. Methods Thirty-two healthy Wistar rats weighting(200 ±25 ) g( male and female were equal) were used to establish the model of heart failure by injecting Isoproterenol. Twenty-four rats model were chosen and randomly divided into 3 groups with 8 rats in each group: heart failure model group,low dosage group(5g·kg-1· d-1) and hige dosage group( 10g·kg-1· d-1). Eight heathy Wistar rats were cho- sen as the control group. The heart failure model group and nornml eontrol group were supplied with equivalent normal saline. All rats of the 4 groups were deliveried by gastric irrigation once a day for 28 days in succession. After 28 days ,24-hour urine volume was measured by metabolism cage,and the renal medullary AQP-2 mRNA expression was detected by reverse transcriptase-poly,nerase chain reaction(RT-PCR). Resuits (1)The 24-hour urine volume of the heart failure model group was obviously decreased compared with the control group ( P 〈 0.05 ) , and the 24-hour urine volume of the high dosage group was obviously increased compared with the heart failure model group ( P 〈 0.05). (2)The kidney medullary. AQP-2 mRNA expression of the heart failure model group was obviously up-regulated compared with the control group( P 〈 0.05 ) , and the expression of high dosage group was obviously down-regulated compared with the heart failure model group( P 〈 0.05 ). There was no statistical significance in the renal medullar3., AQP-2 mRNA expression between the low dosage group and the heart failure model group (P 〉0.05). Conclusion RLF diuretic effect is obvious on rats with chronic congestive heart faihure,and the molecular mechanism may relate to the significantly decreased expression of AQP-2 mRNA in the kidney
