摘 要:目的:探讨二巯丁二钠(DMS)对大鼠铜负荷模型铜代谢的影响。方法:48只Wistar大鼠(均为3个月龄)随机3组,对照组(A组)予正常喂饲;铜负荷组(B组)、治疗组(C组)均同时喂饲含硫酸铜1g/kg的粉状饲料和0.185%硫酸铜水;第4周起C组scDMS300mg/(kg·d)。3组每隔2~4周剪尾取血测Cu,Zn,血清ALT和铜氧化酶;第12周在乙醚麻醉下分别取血、肝、肾及脑组织标本测Cu,Zn含量。结果:A组各种检测值无显著变化;B组血清ALT和Cu,组织Cu和Zn水平均升高,而血清Zn下降;C组治疗后血清ALT和Cu及组织Cu和Zn水平均较B组明显下降,但组织Cu和Zn水平仍明显高于A组。结论:DMS能降低铜负荷大鼠的血清Cu及组织Cu和Zn含量,恢复其肝细胞功能损害。OBJECTIVE: To explore the influence of DMS on copper metabolism in model of copperloaded rats. METHODS :The study was conducted in 48 Wistar rats.They were randomly divided into 3 groups.The control group receives ordinary forage and water,the other 2 groups(copperloaded group and active group)receive forage containing 0.1% copper sultate and water containing 0.185% copper sulfate.The active group was treated with subcutaneous DMS in a dose of 300 mg/(kgd) from the beginning of the fourth week.Blood samples were taken from rats in 3 groups by cutting cauda every third to fifth week and the serum ALT,copper oxidase,Cu,and Zn contents were determined.On 12 weeks samples of blood,liver,kidney,and brain were taken from rats anesthetized with aether and their Cu and Zn contents were determined. RESULTS :No obvious change in metal contents was observed in control group;the levels of serum ALT and Cu and tissue levels of Cu and Zn were increased and serum Zn content was decreased in copperloaded group;and degeneration in the active group the levels of serum ALT and Cu and tissue level of Cu and Zn were lower than that of the copperloaded group but higher than that of the control group. CONCLUSION :The levels of serum Cu and tissue Cu and Zn in copperloaded rats may be reduced by DMS and their hepatocellular function may be restored.
