摘 要:目的观察覆盆子提取物对基质金属蛋白酶(MMP-13)的抑制作用。方法制备覆盆子不同浓度水提物溶液,将基质金属蛋白酶1μL和不同浓度的覆盆子提取物1μL加入到缓冲溶液中,使终体积为50μL;对照组不加入覆盆子水提物溶液;用荧光酶标仪检测荧光强度,计算出不同浓度覆盆子提取物对MMP-13的抑制百分数。结果 IC50=0.04μg.mL-1,覆盆子水提物浓度与MMP-13活性抑制百分数呈正比关系。结论覆盆子水提物体外具有抑制MMP-13活性的作用。Objective To observe the inhibiting effect of Rubus Chingii Hu extract on matrix metalloprotease(MMP-13) in vitro.Methods Obtain crude extract of Rubus Chingii Hu via water extraction,weigh 10 mg of the crude extract to dissolve in distilled water to prepare water extract solution of different concentrations.The 1 μL of MMP-13 and 1 μL different concentration of Rubus Chingii Hu(RCH) extract added to 48 μL of buffer solution,forming final volume of 50 μL.There was not RCH extract in control group.The fluorescence intensity was detected by fluorescence microplate.The inhibiting percentage of Rubus Chingii Hu extract of different concentrations on MMP-13 was calculated.Results IC50=0.04 μg·mL-1.The positive correlation between the concentration of Rubus Chingii Hu extract and the percentage of inhibiting the activity of MMP-13.Conclusion Rubus Chingii Hu extract had inhibiting effect on activity of MMP-13 in vitro.
