摘 要:为初步探讨富勒烯(C60)对人体细胞的潜在危害,以人胚肝细胞(L-02)为研究对象,研究富勒烯暴露对人胚肝细胞的氧化损伤。人胚肝细胞暴露于0、1.25、2.50、5.00、10.00、20.00和40.00μg.mL-1富勒烯悬液24h后,检测细胞内CAT活性和MDA与ROS含量。采用免疫组化法检测SOD蛋白在富勒烯暴露后细胞内的分布情况;Western blotting法观察富勒烯对人胚肝细胞内SOD、GSH-Px和CAT蛋白表达的影响。结果表明,各实验组中细胞内CAT活性均有不同程度的降低,与对照组相比,差异均具有统计学意义(p<0.05);10.00、20.00和40.00μg.mL-1富勒烯暴露组细胞内MDA含量以及10.00μg.mL-1组细胞内ROS的含量明显增加,差异均具有统计学意义(p<0.05)。随着富勒烯含量的增加,细胞内SOD蛋白表达减少且分布不均。同时,富勒烯降低细胞内GSH-Px和CAT蛋白的表达。综合实验结果推测富勒烯引起人胚肝细胞发生氧化损伤可能是通过降低细胞内SOD、GSH-Px和CAT蛋白表达水平来实现的。This study is to determine the potential effects of fullerenes on human body.Treated with 0,1.25,2.50,5.00,10.00,20.00,40.00 μg·mL-1 fullerenes,the oxidative damages of human embryo liver cells were studied.After treated with fullerenes 24 h,the activity of the CAT,and the concentration of MDA and ROS were measured.Cellular protein expression of SOD,GSH-Px and CAT were observed by Western blotting and the protein distribution of SOD was detected by immunohistochemisty.The results indicate that CAT decreased in all groups(p0.05)compared to the control group.Both MDA of 10.00,20.00,40.00 μg·mL-1 groups and ROS of 10.00 μg·mL-1 group increased(p0.05).With increasing the concentration of fullerenes,the expression of SOD,GSH-Px and CAT protein were inhibited.Considering the above results,a conclusion can be drawn that fullerenes inducing oxidative damages on human embryo liver cells may be through lowing down the expression of SOD,GSH-Px and CAT protein.
