摘  要:通过平板对峙法发现菌株P1对马铃薯环腐病菌有很强的抑制作用。经分析,抑菌成分是蛋白质,忍受温度80℃,活性成分在280 nm处有最大吸收峰,分子质量大于10 kDa;通过对菌株形态、生理生化特征以及16S rDNA序列系统发育树分析发现,菌株P1与巨大芽孢杆菌相似度达到100%,确定P1菌株为巨大芽孢杆菌(B.megatherium)。电泳图谱检测该序列由1 391个碱基构成,已在GenBank登录(登录号:FJ823003.1)。Potato ring rot bacteria caused by Clavibacter michiganense subsp.sepedonicum is one of the most important plant diseases.The aim of the experiment is to screen high active antibacterial strain to control this disease and to study the antibacterial substance characteristics.The strain was identified based on the bacteriological characteristics and the 16S rDNA sequence.The culture conditions of P1 strain were optimized by single factor and orthogonal experiment.The strain was identified as Bacillus megatherium.The antibacterial activity substance from P1 could designate as one kind of protein unstable at higher temperature 80 ℃,partly sensitive to proteinase K,chloroform and pH,and stable to trypsin and ultraviolet.Antibacterial substance excreted by Bacillus megatherium P1 is antibacterial protein and it has high activity to CMS-1.The P1 strain could be developed as a promising bio-control bacterium strain.
