摘  要:目的探讨养心颗粒对p53基因转染易损斑块家兔模型血清白细胞介素-18(IL-18)含量的影响。方法以球囊损伤家兔颈总动脉结合高脂饮食喂养8周,8周末在颈总动脉斑块形成处转染重组人p53腺病毒载体制备易损斑块模型。将模型家兔随机分为模型对照组、养心颗粒组、辛伐他汀组,每组6只,另取6只家兔作为空白对照组;给药2周,10周末药物触发斑块破裂,耳缘静脉取血,用ELISA法检测血清IL-18含量。结果与空白对照组相比,各模型组在8周末IL-18含量明显升高(P<0.01);10周末,与模型对照组相比,各治疗组IL-18含量降低(P<0.01);两治疗组比较,养心颗粒组IL-18含量低于辛伐他汀组(P<0.01)。结论养心颗粒能明显降低p53基因转染易损斑块家兔模型血清IL-18含量,这可能为其稳定易损斑块的作用机制之一。Objective To study the effect of Yangxin granule on serum interleukin 18(IL-18) in vulnerable plaque of rabbit model with p53 gene transfection.Methods Carotid artery wall injuries were induced by intravascular balloon in rabbits.At the end of the 8th week,recombinant adenovirus vector by transfecting p53 gene were injected to plaques of carotid segments.Rabbits were randomly divided into four groups:Model control group,Yangxin granule group,simvastatin group and blank group(6 in each group).Following of administration for two weeks,plaques were ruptured because of pharmacological triggering.At the end of the 8th and 10th weekend,the contents of serum IL-18 was detected by ELISA method.Results Compared with blank group,the content of serum IL18 in other groups was obviously increased at the end of the 8th week(P0.01).Compared with model control group,the content of serum IL-18 in treatment groups was declined at the end of 10th week.There was significant difference between Yangxin granule group and simvastatin group(P0.01).The content of serum IL-18 in Yangxin granule group was lower than that in simvastatin group(P0.01).Conclusion Yangxin granule could remarkably decrease the content of serum IL-18 in vulnerable plaque of rabbit model with p53 gene transfection,which might be one of the mechanisms for stabilizing vulnerable plaque.
