摘  要:目的:新生兔骨髓间充质干细胞(BMSCs)的培养方法,从各方面鉴定体外培养BMSCs的生物学特性。方法:采用贴壁筛选法纯化MSCs,镜下连续细胞的形态学观察。从形态学特征、表面标志物及其具有的向骨、脂肪分化的功能等多方面经行综合鉴定。结果:原代BMSCs呈集落状生长,细胞呈梭形、呈放射状排列。传代后大都均匀分布生长,细胞生长迅速;可向成骨细胞、脂肪细胞分化。结论:贴壁筛选法可成功分离和培养新生兔的BMSCs,BMSCs能向成骨细胞、脂肪细胞分化,可以作为组织工程中种子细胞的来源。Objective:To establish a method of cultivating rabbit bone marrow mesenchymal stem cells(MSCs)in vitro and the biological characteristics thereof from many ways.Methods:Rabbit BMSCs were isolated by cultured with L-DMEM.The mor-phology of BMSCs was observed in passage culture under inverted microscope and identification from morphology,surface antigens and differentiation.Results:Cultured primary BMSCs were spindle shaped and had a typical fibrob lastlike morphology,proliferated in colonies.After a series of passages,the attached cells became morphological homo-geneous.Conclusion:The bone marow MSCs can be successfully isolated,cultured and purified from rabbits BMSCs through L-DMEM,could make identification into bone cells and fat cells.The BMSCs can be used as a potential cell source for tissue engineering.
