身痛逐瘀汤对骨癌痛小鼠痛行为及脊髓星形胶质细胞活化的影响

摘　　要：目的探讨身痛逐瘀汤的镇痛作用及对脊髓水平胶质细胞纤维酸性蛋白(GFAP)表达的影响。方法 将C3H/HeNCrlVr雄性小鼠100只随机分为正常组(0d,8只)、假手术组(30只)、模型组(30只)、中药1组(8只)、中药2组(8只)、中药3组(8只)和溶媒组(8只)。中药1、2、3组和溶媒组分别给予身痛逐瘀汤水煎浓缩制剂0.1、0.3、0.9g生药/0.4mL和0.4mL生理盐水灌胃,于小鼠骨癌痛造模第14天开始,每天灌胃1次,共7天。用热辐射刺激仪测定各组小鼠行为学指标热痛缩足潜伏期(PWTL)。第21天行为学测试后,取脊髓腰膨大部位,分别应用Real-time quantitative RT-PCR和Western blot检测GFAP mRNA及蛋白的表达。结果 与正常组[0d,PWTL:(15.91±1.65)s]和假手术组[PWTL:4d:(13.33±1.44)s;7d:(11.28±0.61)s;10d:(15.47±2.46)s;14d:(15.69±1.98)s;21d:(15.69±1.68)s]比较,模型组小鼠的PWTL值[4d:(13.24±1.02)s;7d:(11.30±1.09)s;10d:(9.12±0.54)s;14d:(7.79±0.77)s;21d:(6.36±0.59)s]随着时间的推移进行性下降(P<0.05),而脊髓水平GFAP mRNA及蛋白表达逐渐增加。与模型组比较,溶媒组及中药1、2、3组之间在14d时间点PWTL值差异无统计学意义(P>0.05)。在21d时间点,与模型组和溶媒组比较,中药2组、3组小鼠PWTL升高(P<0.05),脊髓组织GFAP mR-NA及蛋白表达水平降低(P<0.05),而模型组、溶媒组、中药1组之间PWTL值及GFAP表达水平差异无统计学意义(P>0.05)。结论 身痛逐瘀汤具有镇痛作用,抑制脊髓水平星形胶质细胞的增殖活化可能是其机制之一。Objective To study the analgesic effect of Shentong Zhuyu Decoction（SZD） and its effect on the expression of the spinal cord glial fibrillary acidic protein（GFAP）.Methods One hundred C3H/HeNCrlVr male mice were randomly divided into the normal group（n=8）,the sham operation group（n=30）,the model group（n=30）,the Chinese medicine（CM） group 1（n=8）,the CM group 2（n=8）,the CM group 3（n=8）,and the vehicle group（n=8）.0.1 g crude drug of SZD/0.4 mL,0.3 g crude drug of SZD/0.4 mL,0.9 g crude drug of SZD/0.4 mL,and 0.4 mL normal saline were respectively given by gastrogavage to mice in CM 1,2,3 groups and the vehicle group,once daily for seven days starting from Day 14.The paw withdrawal thermal latency（PWTL）,as the behavior indicator,was assessed in mice using radiant thermal stimulator.The lumbar enlargement of the spinal cord was taken after the behavioral test on Day 21.GFAP mRNA and protein expressions were detected using Real-time quantitative RT-PCR and Western blot.Results Compared with the normal group（Day 0）（PWTL：15.91 ± 1.65 s） and the sham operation group（PWTL：Day 4：13.33±1.44 s;Day 7：11.28±0.61 s;Day 10：15.47±2.46 s;Day 14：15.69±1.98 s;Day 21：15.69±1.68 s）,the PWTL value in the model group（Day 4：13.24±1.02 s;Day 7：11.30±1.09 s;Day 10：9.12±0.54 s;Day 14：7.79 ± 0.77 s;Day 21：6.36±0.59 s） progressively decreased（P0.05） as time went by,while the spinal cord GFAP mRNA and protein expressions gradually increased.Compared with the normal group（Day 0） and the sham operation group（Day 14）,the PWTL value in the CM groups and the vehicle group obviously decreased on Day 14（P0.05）.The PWTL value was not significantly different among the model group,CM groups,and the vehicle group on Day 14（P0.05）.On Day 21 the PWTL value of CM group 2 and 3 increased and the spinal cord GFAP mRNA and protein expression levels decreased when compared with the model group and the vehicle group（P0.05）.But no significant diffe
