水合氯醛对小鼠生长发育、学习记忆和治疗指数的影响

摘　　要：目的水合氯醛对小鼠生长发育、学习记忆和治疗指数的影响。方法用各组小鼠体重变化趋势观察水合氯醛对小鼠生长发育的影响,避暗法和跳台法观察水合氯醛对小鼠学习记忆的影响,用分组法测量水合氯醛对小鼠催眠的ED50、LD50和治疗指数。结果在跳台和避暗实验中,与NS组相比,第一天C1、C2、C3(水合氯醛剂量分别为0.005、0.0075、0.01ml·10g-1)潜伏期缩短(P<0.01),避暗实验C1、C3组和跳台实验C2、C3组错误次数增加(P<0.01),第二天,与NS组相比,避暗实验中C1、C3组错误次数增加(P<0.05或P<0.01),C1组潜伏期缩短(P<0.05),第三、四天未见显著性差异(P>0.05);各组小鼠在1-5d体重大致呈增长趋势,组间比较无明显差异。结论水合氯醛可在短期内损害小鼠学习记忆能力,对生长发育无明显影响,治疗指数为3.842,用药安全范围较小。Objective：To investigate the effects of chloraldurate on the growth,learning-memory and therapeutic index of mice.Methods：The experiments were designed to study the effects of chloraldurate on the growth by measuring the weight of mice,learning-memory by step-through and step-down test,the hypnosis ED50,LD50 and therapeutic index of chloraldurate on mice by grouping method.Results：In the step-dawn and step-through tasks,as compared with the NS group,the step-down latency decreased in the C1,C2 and C3 groups（the dose of chloraldurate are 0.005,0.0075 and 0.01 mL/10g） at the first day（P 0.01）,the error times of C1,C3 groups in step-through test and C2,C3 groups in step-down test increased（P 0.01）.At the second day,in the step-through test,the error times of C1 and C3 groups increased（P 0.05）,and the step-down latency of C1 group decreased（P 0.05）.There has no obvious difference at the third and fourth day（P 0.05）.The weight of all groups showed the tendency of increase from the first day to the fifth day,and there is no obvious difference among them.Conclusion：Chloraldurate has no obvious effect on the growth of the mice,but may temporarily impair the learning-memory.The therapeutic index of chloraldurate is 3.842,and the scope of drug safety is low.
