摘 要:采用PCR-SSCP技术检测山羊GDF9基因外显子突变位点,发现编码序列第183 bp、719 bp、959 bp、1189bp处分别发生了C→A、C→T、A→C、G→A的单碱基突变。序列分析显示C183A突变没有引起氨基酸的改变,C719T突变使蛋白质第240位氨基酸残基由缬氨酸变为丙氨酸,A959C突变使第320位氨基酸残基由谷氨酰胺变为脯氨酸,G1189A突变导致第397位氨基酸残基由缬氨酸变为异亮氨酸。应用LDR技术对济宁青山羊(234只)、鲁北白山羊(90)只、沂蒙黑山羊(84只)进行GDF9四个突变位点的群体检测,利用SAS软件GLM过程对四个位点的基因型效应和产羔数进行了最小二乘分析,结果表明C183A突变对济宁青山羊和鲁北白山羊产羔数没有显著影响,但对沂蒙黑山羊的产羔数有显著影响(P<0.05),CC型产羔数比AC型多0.11只(P<0.05);C719T突变和A959C突变对济宁青山羊、鲁北白山羊、沂蒙黑山羊的产羔数均没有显著影响(P>0.05);G1189A突变对山羊产羔数没有显著影响(P>0.05),对鲁北白山羊产羔数有显著影响(P<0.05),AG型产羔数比AA型多0.42只(P<0.01),G1189A突变对沂蒙黑山羊产羔数有极显著影响(P<0.01),AA型产羔数分别比AG型、GG型多0.34只(P<0.05)、0.38只(P<0.05)。The mutations in the GDF9 gene exon of goat were detected using PCR-SSCP.The result showed four splice mutations in the coding sequence of GDF9(183bp C→A,719bp C→T,959bp G→A and 1189bp G→A).C183A mutation couldn't change amino acid.C719T mutation caused valine in 240 amino acid to turn into alanine.A959C mutation caused glutamine in 320 amino acid to turn into proline.G1189A mutation caused valine in 397 amino acid to turn into isoleucine.Mutation detection and reproduction trait was analyzed through LDR to Jining grey goat(234) and Lubei white goat(90) and Yimeng black goat(84).The res lts showed that there was no significant difference effect of mutation in C183A point for litter size to Jining grey goat and Lubei white goat,but Yimeng black goat at P=0.05 level.The does with genotype CC had 0.11(P0.05) kids more than that with genotype AC in Yimeng black goat(P0.05).There was no difference in C719T and A959C points for litter size to Jining Grey goat and Lubei white goat and Yimeng black goat.There was no significant difference effect of mutation in G1189A point for litter size to Jining grey goat,but Lubei white goat at P=0.05 level and Yimeng black goat at P=0.01 level.The does with genotype AG had 0.42(P0.01) kids more than that with genotype AA in Lubei white goat,and the does with genotype AA had 0.34(P0.05) or 0.38(P0.05) kids more than that with genotype AG or GG in Yimeng black goat,respectively.
