摘 要:目的:研究促甲状腺激素(TSH)β基因剪接变体在BALBC小鼠垂体、甲状腺、骨髓、脾、外周血和淋巴结中的表达情况。方法:选取雄性6~8周龄BALBC小鼠6只,提取小鼠垂体、甲状腺、骨髓、脾、外周血和淋巴结的总RNA,用DNA酶处理后,应用逆转录聚合酶链反应(RT-PCR)方法检测TSHβ剪接变体的表达情况。结果:垂体、甲状腺、骨髓、脾、外周血和淋巴结中均可见TSHβ剪接变体的特异性表达,且在垂体和外周血中的表达量较高。测序证实该剪接体含有部分第4内含子和整个第5外显子的编码区,没有第4外显子。结论:TSHβ剪接变体在BALBC鼠的垂体、甲状腺、骨髓、脾、外周血和淋巴结中均有表达。Objective:To determine whether the novel thyroid stimulating hormone β-subunit (TSHβ) splice variant was expressed in pituitary, thyroid tissues,bone marrow(BM), spleen, peripheral blood and lymph nodes of BALB C mice. Methods:Six male BALB C mice (6-8 week-old) were selected in this study. Total RNA was isolated respectively from pituitary, thyroid tissues, BM, spleen, peripheral blood leukocytes (PBL) and lymph nodes of BALB C mice. The samples were treated with an RNase-free DNase. Reverse transcriptase polymerase chain reaction (RT-PCR) was performed to detect the expression of TSHβ splice variant in these tissues. Results:The TSHβ splice variant was expressed in pituitary, thyroid tissues, BM, spleen, PBL and lymph nodes in BALB C mice. The level of the TSHβ splice variant expression was higher in pituitary and PBL than that of other tissues.?The sequence analysis confirmed that it consisted of a portion of intron 4 and all of the coding regions of exon 5, but none of exon 4 codon. Conclusion:The TSHβ splice variant is expressed in pituitary, thyroid tissues, BM, spleen, PBL and lymph nodes in BALB C mice.
