苣荬菜乙醇提取液抗贝氏柯克斯体的初步研究

摘  要:目的初步探讨苣荬菜(北败酱)乙醇提取物的抗Q热病原菌-贝氏柯克斯体药效。方法用乙醇提取苣荬菜,1g干生药获得1mL提取液;用贝氏柯克斯体腹腔感染Balb/c小鼠(105菌体/只)感染后第2d用倍比(5-160倍)稀释苣荬菜乙醇提取液腹腔给药,每天1次、连续给药5d;第7d处死小鼠并取小鼠脾脏组织提取DNA,用种特异性荧光定量PCR检测DNA。结果 10、20、40、80倍稀释药物组小鼠脾脏贝氏柯克斯体载量约2~5倍少于非药物处理对照组,其中20倍稀释药物组的小鼠脾脏贝氏柯克斯体载量显著(5倍)少于对照组。结论我们的研究首次证明苣荬菜乙醇提取物对贝氏柯克斯体体内生长有明显的抑制作用,提示它具有潜在的抗贝氏柯克斯体药效。Sonchus arvensis L is one of the herbal medicines that were used to treat certain infectious diseases in Chinese medicine.In this study,the ethanol solution extracted from Sonchus arvensis L(1 mL solution extracted from 1 gram of dried herb) was applied to treated mice infected with C.burnetii,a pathogen of Q fever.From day 2 to 6 after the intraperitoneal inoculation of C.burnetii(105 organisms/ per mouse),the 5-,10-,20-,40-,and 80-fold diluted solutions were intraperitoneally inoculated into mice(0.5 mL/ per mouse) once per day,respectively.The mice were sacrificed and their spleen tissues were collected for preparing DNA.The burdens of C.burnetii in the mouse spleens were detected by quantitative polymerase chain reaction.The results exhibited that the coxiella burdens of mice treated with 10-,20-,40-,or 80-fold diluted solutions were 2-to 5-fold lower than that of untreated mice,and that of mice treated with 20-fold diluted solutions were significantly(5-fold) lower than that of untreated mice.Our results firstly confirm that the ethanol extracts of S.arvensis L has the capability to inhibit the growth of C.burnetii in bodies and suggest that it possesses a potential drug's effect against C.burnetii,an obligate intracellular bacterium.
