“决明山楂燕麦胶囊”的毒理学试验

摘　　要：目的观察决明山楂燕麦胶囊的急性及长期毒性反应,以确保用药安全性。方法急性毒性反应以10 g/kg(小鼠最大耐受量)的决明山楂燕麦给小鼠一次性灌胃,连续观察14 d,逐日记录毒性反应、死亡数,并作病理学检查。长期毒性反应以低剂量组1.6 g/kg、高剂量组2 g/kg给大鼠连续灌服8周,对大鼠进行一般状况、血液学指标、血液生化指标、主要脏器指数和组织病理学观察和检查。结果急性毒性实验未见小鼠出现毒性反应及死亡。长期毒性结果显示,低、高剂量组大鼠的一般状况、体质量、血液学指标、血液生化学指标及主要脏器系数与对照组比较无显著性差异;肉眼观察服药组心、肝、脾、肺、肾外观形态、颜色均无异常改变,光镜检查也未见各脏器组织结构和细胞形态异常。结论决明山楂燕麦对实验动物安全无毒,这可为临床应用提供有用信息。Objective To ensure the safety of drug use,the acute and Long-period toxic reactions of semen cassiae hawthorn oat were observed.Methods For the acute toxic reactions,the semen cassiae hawthorn oat was orally given for one-time at 10 g/kg dosages（the maximum tolerance dose in mices）,observed continuously for 14 days.Daily record toxicity,deaths and pathology examination were carried out.For the long-period toxic reactions,the semen cassiae hawthorn oat was orally given for 8 weeks continuously at low（1.6 g/kg） and high（2 g/kg） dosages.The rats＇ general physical state,hematological indexes,biochemical indexes,organ coefficients and histopathological examination were carried out.Results There were no the toxic reaction and animals death at the acute toxic experiments.For the long-period toxic reactions,there were no significant differences of the general state,weight changes,hematological indexes,biochemical indexes and organ coefficients in rats of low and high dosage group when compared with control group.Macroscopic observation of heart,liver,spleen,lung and kidney showed no abnormal change of appearance shape and color.Pathologic examination did not show any structural and cellular abnormalities of each organ.Conclusions semen cassiae hawthorn oat is safe and non-toxicit for experimenta animals,these data may provide useful information for clinical use.
