摘  要:探讨重组水蛭素Ⅲ(rHV3)对半乳糖损伤的人晶状体上皮细胞(hLEC)热应激蛋白(Hsp)表达的影响。采用125×10-3mol/LD-半乳糖建立人晶状体上皮细胞SAR01/04损伤模型,用重组水蛭素Ⅲ干预受损细胞,设置正常组、模型组、和重组水蛭素Ⅲ组,用倒置显微镜观察各组细胞密度和形态,RT-PCR检测人晶状体上皮细胞内3种热应激蛋白mRNA的表达。结果显示,125×10-3mol/LD-半乳糖导致人晶状体上皮细胞显著损伤,3种热应激蛋白,即Hsp70、Hsp27和αB-晶体蛋白的mRNA表达显著上调。重组水蛭素Ⅲ的干预使细胞形态趋于正常,细胞内Hsp70、Hsp27和αB晶体蛋白的mRNA表达显著下调。因此,重组水蛭素Ⅲ能有效缓解高浓度半乳糖导致的热应激蛋白应激耐受,从而保护晶状体上皮细胞。The aim of the paper is to investigate the effects of recombinant hirudin variant Ⅲ(rHV3) on the expression of heat shock proteins in human lens epithelial cell(hLEC) line SAR01/04 damaged by galactose.Damaged hLEC model was established using 125×10-3mol/L D-galactose,and rHV3 was used to interfere the damaged cells.There were three experimental groups: normal group,model group,and rHV3 group.The cells were observed under the phase-contrast microscope for any morphological changes.The expression of three heat shock proteins at the mRNA levels in cells was detected using RT-PCR.The cells were significantly damaged,and the mRNA levels of Hsp70、 Hsp27 and αB-crystallin were upregulated,under the condition of 125×10-3mol/L D-galactose.rHV3 prevented the cellular morphological changes and elevation of Hsp70、 Hsp27 and αB-crystallin mRNAs.These results indicated that rHV3 could attenuate stress tolerance of heat shock proteins insulted by D-galactose and have the positive function in protecting the hLEC.
