摘 要:目的观察IgA肾病(IgA nephropathy,IgAN)患者与正常人的血清热聚合IgA1(aggregated IgA1,aIgA1)对足细胞及系膜细胞增殖的影响。方法利用层析法获得IgAN患者与健康人的血清单体IgA1(monom eric IgA1,m IgA1),将m IgA1热聚合为aIgA1,分别刺激小鼠MPC5足细胞及MSC-1097系膜细胞株,利用MTT法检测细胞增殖。结果①不同浓度aIgA1刺激组间足细胞MTT吸光度差异有统计学意义。相比于阴性对照组(SFM),1 mg/m l、2 mg/m l组吸光度分别为0.456(P=0.033)、1.663(P=0.010)。②不同浓度aIgA1刺激组间系膜细胞MTT吸光度差异有统计学意义。相比于阴性对照组(SFM),0.25 mg/m l、0.5 mg/m l、1 mg/m l组吸光度分别为1.667,1.736,1.772(P=0.000),2 mg/m l组吸光度为1.148(P=0.086)。结论足细胞可能会直接对IgA1产生反应;aIgA1可以影响足细胞及系膜细胞增殖,且对两种细胞增殖的影响存在不同。Objective To explore the effects of serum aggregated IgA1(aIgA1)on the proliferation of podocyte and mesangial cell in patients with IgA nephropathy(IgAN) and healthy controls.Methods Blood samples of patients with primary IgAN and healthy controls were collected.Serum monomeric IgA1(mIgA1) was isolated using a chromatography,then heated to aggregated form(aIgA1).Mouse podocyte(MPC5) and mesangial cell(MSC-1097) were cultured in vitro,then the effects of aIgA1 on proliferation of the two kinds of cells were observed by MTT method.Results ①There was a significant difference in the MTT absorbency of podocyte cells stimulated by different concentration of aIgA1(P=0.000);as compared to the control(SFM),the absorbency of 1 mg/ml group and 2 mg/ml group was 0.456(P=0.033)and 1.663(P=0.010),respectively.②There was a statistical difference in the MTT absorbency of mesangial cell stimulated by different concentration of aIgA1(P=0.000);as compared to the control(SFM),the absorbency of 0.25 mg/ml group,0.5 mg/ml group and 1 mg/ml group were 1.667,1.736 and 1.772(P=0.000),respectively;while it was 1.148 in the 2 mg/ml group(P=0.086).Conclusion The podocyte can response to IgA1 in a direct manner;aIgA1 mat play an important role on the proliferation of podocyte and mesangial cells by a different way
