摘  要:采用3种浓度的EMS对离体培养的文心兰类原球茎薄切片进行不同时间的诱变处理研究。发现EMS使部分薄切片褐化死亡,再生类原球茎的生长受到抑制,再生苗数量减少;EMS的伤害作用随诱变剂浓度的提高和处理时间的延长而加重,但对试管苗的生长影响并不大。采用10条RAPD引物进行的PCR检测结果表明,经EMS处理的再生苗DNA序列发生变异,表现出RAPD图谱带型的多态性。不过,EMS浓度提高到0.4%后,多态率的升高不明显,EMS合适的诱变剂量为0.4%浓度,处理时间为2~4 d。Pieces of thin cell layers(TCLs) of Oncidium protocorm-like bodies(PLBs) were treated in vitro with EMS for the induction of mutation and it was found that mutagen EMS had some TCLs got browning and died.The regenerated PLBs grew slowly and the number of regenerated plantlets were reduced.There were obvious injured effects on PLBs,but less injured effects on plantlets.The injured effects got more and more serious with higher concentration of mutagen and longer time treatment.The results of 10 primers of RAPD reaction indicated that the molecule maps of RAPD had some polymorphisms which mean the sequence of DNA had changed.The ratio of polymorphisms got higher with higher concentration of mutagens and longer time treatment.However,the ratio of RAPD polymorphisms had no obvious raised when the concentration of EMS reached to 0.4%.The concentrations of 0.4% EMS treatment are suitable dosage for Oncdium chemical induction in vitro.
