摘 要:以4种脱水蔬菜为对象,研究ATP生物发光法检测辐照处理脱水蔬菜样品的含菌量时,ATP生物发光强度与细菌含量不一致的原因。结果表明:辐照对样品的本底发光无明显影响,也未改变荧光素酶发光系统的光谱;电离辐射在杀灭样品中细菌的同时降低了细菌体内ATP酶的活性,使死亡细菌中的ATP不能随细菌的死亡而降解,并从死亡细菌体内扩散出来,从而增加了样品中游离ATP的浓度,导致细菌ATP提取液的发光强度与实际含菌量不一致;去除样品中游离ATP后,细菌ATP提取液的发光强度与实际含菌量的动态变化已趋于一致。ATP生物发光技术可很好地应用于辐照产品的细菌检测。ATP bioluminescence intensity of 4 kinds of irradiated dehydrated vegetables was inconsistent with the bacteria number,the reasons were investigated in this paper.Results showed that irradiation had little effect on background luminescence,and there was no effect on luciferase-luminous system.When irradiation killed the bacteria,the ATPase activity also decreased.As a result,the ATP content in bacteria didn’t decreased with the killed of bacteria,which contributed to the increase of free ATP in ATP extract and finally led to the disagreement between the bioluminescence intensity and the actual number of bacteria.When the free ATP in the dehydrated vegetable was removed,the bioluminescence intensity of ATP extract was consistent with the actual number of bacteria in irradiated dehydrated vegetable and ATP bioluminescence technology could be used in bacteria detection of irradiated samples.
