摘  要:[目的]比较栽植的不同生长年限膜荚黄芪中黄酮类成分的高效液相色谱信息、毛蕊异黄酮和芒柄花素的含量以及总黄酮含量.[方法]采用HPLC-DAD方法比较不同生长年限膜荚黄芪中黄酮类成分以及毛蕊异黄酮和芒柄花素的含量,色谱柱为BDS Hypersil C18柱(250.0 mm×4.6 mm,5μm,Thermo),应用二极管阵列检测器,流动相为乙腈-水,梯度洗脱50 min,流动速度为1.0 mL/min,检测波长为254 nm;采用比色法测定总黄酮含量.[结果]不同生长年限膜荚黄芪中黄酮类成分指纹图谱有7个主要共有峰,在概貌上基本一致,但出峰数量和相对峰面积有所差别;1,2,3年生膜荚黄芪中总黄酮含量分别为1.02,1.23,1.79 mg/g,毛蕊异黄酮为0.337,0.121,0.119 mg/g,芒柄花素为0.101,0.031,0.032 mg/g.[结论]不同生长年限膜荚黄芪中黄酮类成分含量随着年限的增长而增高,黄酮类成分的种类基本保持不变.OBJECTIVE To compare the high pressure/performance liquid chromatography(HPLC) information of flavonoids,calycosin and formononetin content and total flavonoid content in different growth years of Astragalus membranaceus.METHODS The flavonoids in Astragalus membranaceus and the contents of calycosin and formononetin were compared by using the HPLC and diode array detector(DAD).The chromatographic column was BDS Hypersil C18 column(250.0mm×4.6mm,5μm,Thermo),and using DAD,mobile phase was acetonitrile water,gradient elution was 50min,flow rate was 1.0mL/min,detection wavelength was 254nm,and the content of total flavonoid was determined by colorimetric technique.RESULTSThere were seven main common peaks in the finger print of flavonoids in different growth years of Astragalus membranaceus,and the pattern in the profile was basically the same,but the relative peak areas were different.In 1,2 and 3 growth years of astragalus membranaceus,the total flavonoids were 1.02,1.23 and 1.79mg/g,calycosins were 0.337,0.121 and 0.119mg/g,and formononetins were 0.101,0.031 and 0.032mg/g,respectively.CONCLUSION The content of flavonoid in different growth years of Astragalus membranaceus is increased along with the growth period,and the types of flavonoids remain coincidence.
