大黄蜇虫丸影响免疫性肝纤维化大鼠TIMP-1表达的动态观察

摘　　要：目的观察大黄蜇虫丸对免疫性肝纤维化大鼠TIMP-1表达影响的动态变化,探讨大黄蜇虫丸抗肝纤维化的作用机制。方法将120只健康雄性SD大鼠随机分为6组。即正常组,模型组,大黄蜇虫丸低剂量组,大黄蜇虫丸中剂量组,大黄蜇虫丸高剂量组,秋水仙碱组,各20只,除正常组外其它各组均注射人血清白蛋白制作免疫性肝纤维化大鼠模型,模型制作完成后,各组采用相应的药物连续干预4周,分别于药物治疗后1、2、3、4周末随机处死5只大鼠,用酶联免疫吸附法检测血清中基质金属蛋白酶抑制因子-1(TIMP-1)的含量变化。结果①模型组随着周次的延长血清TIMP-1同步增高,差异有统计学意义(P<0.01);②大黄蜇虫丸低剂量组、大黄蜇虫丸中剂量组、大黄蜇虫丸高剂量组随着周次的延长血清TIMP-1同步降低,差异有统计学意义(P<0.01),其中大黄蜇虫丸中剂量组优于其他两组(P<0.01);③大黄蜇虫丸有明显的抑制肝纤维化模型大鼠肝组织TIMP-1表达的作用,且抑制作用要优于秋水仙碱组(P<0.01)。结论大黄蜇虫丸可有效防治大鼠免疫性肝纤维化,其作用机制可能是通过抑制肝纤维化组织TIMP-1的表达而实现。Objective To observe the dynamic changes of the effect of Dahuang Zhechong Pill（DZP） on the expression of TIMP-1 in rats with immune hepatic fibrosis and study the mechanism of the pill for anti-hepatic fibrosis.Methods 120 healthy male SD rats were randomly divided into six groups：normal group,model group,and groups of low,medium,high dose of DZP,and colchicines group,20 rats each group.Rats in all the other groups were made models of immune hepatic fibrosis induced by human serum albumin except the normal group.All groups were given predetermined treatment for four weeks.Five rats were randomly sacrificed each time at the 1st,2nd,3rd weekend after treatment to detect their serum content of TIMP-1 by ELISA.Results ①The results showed that of the serum levelof TIMP-1 increased significantly of model group（P〈0.01）.②The results showed that of the serum level of TIMP-1 decreased significantly of DAZCP low-dose group,medium-dose group and high-dose group（P〈0.01）,The medium-dose group is good for other groups（P〈0.01）.③Dahuang zhechong pill inhibited obviously the expression of TIMP-1 in hepatic tissue of rat with hepatic fibrosisin comparison with that in colchicines.Conclusion DZP can prevent immune hepatic fibrosis in rats,probably by inhibiting the expression of TIMP-1 in hepatic tissue.
