摘  要:CBLs(calcineurin B-like proteins)是植物细胞中1个重要的Ca2+传感蛋白家族,它们广泛参与植物对高盐和干旱等非生物逆境的应答反应。在玉米基因组中存在至少9个CBL编码基因,其中ZmCBL5因为存在可变剪接而有编码框为657bp和444bp两种转录本,我们称之为ZmCBL5-1和ZmCBL5-2。本论文运用RT-PCR方法克隆了ZmCBL5-2的编码框cDNA,然后构建了其植物表达载体并转化了拟南芥,经PCR检测证明获得T1代转基因植株,为进一步研究其在抗逆性中的作用奠定了基础。The CBLs ( calcineurin B - like proteins) are an important family of calcium sensors in plant cell. They participate in the responses of plants for abiotic stresses, like high salinity and drought. There are at least 9 CBL genes in the genome of maize. Among them,ZmCBL5 has two transcripts beccause of the alternative splicing. The transcripts harbor the open reading frames of 657bp and 444bp respectively and were designated as ZmCBL5 - 1 and ZmCBL5 - 2. In our study, the complete coding region of eDNA for ZmCBL5 - 2 was cloned by RT - PCR and was constructed into plant expression vector. Then the construction was introduced into Arabidopsis. The T1 transgenic plants were identified by PCR. The results laid a foundation for the resistance of ZmCBL5 to stress.
