人类骨髓基质细胞系免疫组化标本的制备

摘　　要：采用体外培养技术制作人类骨髓基质细胞系，在８ｃｍ２平皿中制备原代和传代细胞标本，用ＡＢＣ法在皿中直接行Ｖｉｍｅｎｔｉｎ、Ｄｅｓｍｉｎ、αＳｍｏｏｔｈＭｕｓｃｌｅＡｃｔｉｎ单克隆抗体和ＶｏｎＷｉｌｅｂｒａｎｄＦａｃｔｏｒ抗体染色。结果显示：平皿中细胞分布均匀，无污染、破损和脱片，方法简易，优于盖玻片法，可用于贴壁粘附细胞的免疫组化研究。To improve the effect and efficiency of immunocytochemical staining,we prepared the specimens of human bone marrow stromal cell lines with in vitro culture technique in 8.0 cm2 petri dishes and performed the immunocytochemical staining for Vimentin,Desmin,Smooth Muscle Actin and Von Willebrand Factor directly in these dishes by the standard ABC method.The results showed that the cells grew well and distributed evenly in these dishes without any infection and that the staining effect was reliable without any detachment.In conclusion,this simple and effective way is superior to that one using coverslip(s),and allows immunocytochemical study for any adherent cells.
