摘 要:目的观察葱白提取物及其不同制剂对急性心肌缺血兔血清一氧化氮(NO)和肌钙蛋白T(TnT)的影响,探讨葱白提取物纳米乳口腔喷雾剂治疗急性心肌缺血的作用机制。方法选用体重为2.5~3.0 kg的健康雄性日本大耳白兔48只,随机分为假手术组、空白模型组、硝酸甘油组、葱白提取物组、葱白纳米乳组和葱白纳米乳原位凝胶组,每组8只。制备急性心肌缺血模型,术后及时给药,假手术组和空白模型组舌下给予同体积的生理盐水0.5 ml,各治疗组分别给予相应的药物0.5 ml。给药后观察3 h,打开腹腔,腹主动脉取血,测定血清NO、TnT的含量。结果葱白提取物组、葱白纳米乳组、葱白纳米乳原位凝胶组血清中NO水平均显著升高(P〈0.05),血清中TnT水平均明显降低(P〈0.05);硝酸甘油组、葱白提取物组、葱白纳米乳组、葱白纳米乳原位凝胶组之间比较,差异无统计学意义。结论葱白提取物及其纳米乳和纳米乳原位凝胶均能促进血管舒张因子的分泌而改善心肌缺血,但是不同制剂之间差异无统计学意义,其原因还有待进一步深入研究。Objective To observe the influence of fistular onion stalk extract and its different preparations on serum nitric oxide(NO)and cardiac troponin T(TnT)levels in acute myocardial ischemia rabbit,and probe the mechanism of oral nanoemulsion spray of fistular onion stalk extract in the treatment of acute myocardial ischemia.Methods 48 healthy male Japanese white rabbits weighing 2.5—3.0 kg were randomly divided into sham operation group,model group,nitroglycerin group,fistular onion stalk extract group,fistular onion stalk nanoemulsion group and fistular onion stalk nanoemulsion gel group,with 8 rabbits in each group.After preparation of acute myocardial ischemia,the sham operation group and the model group were administered sublingual saline(0.5 ml),sublingual nitroglycerin group was given nitroglycerin suspension(0.5 ml).The other three groups were given fistular onion stalk extract and its nano dose sublingual formulation(0.5 ml).After administration the drugs for 3 h,serum NO and TnT levels were observed.Results Serum NO levels in fistular onion stalk extract group,fistular onion stalk nanoemulsion group and fistular onion stalk nanoemulsion gel group increased significantly(P0.05)and serum TnT levels in those groups decreased obviously(P0.05).There is no significant difference in both NO and TnT levels among nitroglycerin group,fistular onion stalk extract group,fistular onion stalk nanoemulsion group and fistular onion stalk nanoemulsion gel group.Conclusion Both fistular onion stalk extract and its in situ nanoemulsion and nanoemulsion gel preparations could promote the secretion of vascular relaxing factor and then improve the myocardial ischemia.However there was no significant difference between the different preparations,the reason remains to be further study.
