温阳活血化痰法对心力衰竭大鼠心功能及肿瘤坏死因子-α表达的影响

摘　　要：目的:研究温阳活血化痰法对异丙肾上腺素(ISO)诱导的充血性心力衰竭(CHF)大鼠心功能及心肌肿瘤坏死因子-α(TNF-α)蛋白表达的影响,探讨TNF-α在CHF发病中的作用及该法防治CHF心肌重塑可能的作用机理。方法:采用ISO皮下注射法制备CHF模型,药物治疗6周后,进行血流动力学及病理形态学检测,采用免疫组化法测定心肌TNF-α蛋白表达变化。结果:温阳活血化痰法能提高CHF模型大鼠dp/dtmin、dp/dtmax和LVSP水平、降低LVEDP水平;降低心脏重量指数,抑制CHF模型大鼠心肌上调的TNF-α蛋白表达。结论:温阳活血化痰法能明显改善CHF大鼠心肌收缩和舒张功能,抑制细胞重塑,可能与其下调TNF-α蛋白表达有关。Objective：To investigate the effect of TCM treatment ＂warming Yang promoting blood circulation and resolving phlegm＂ on Tumor necrosis factor-alpha（TNF-α） expression in congestive heart failure（CHF） rat model induced by isoproterenol（ISO）,and to explore the role of TNF-αin CHF and the therapeutic mechanism of the treatment on myocardial remodeling.Method：CHF animal models were induced by ISO.After 6 weeks,hemodynamics and pathological morphology were assayed and the expression of TNF-αprotein were detected by immunohistochemistry.Results：The TCM treatment could increase the level of dp/dtmax,dp/dtmin and LVSP,and down-regulate LVEDP,and lower the heart weight index.It also could inhibit TNF-α protein expression in CHF.Conclusion：The treatment can improve myocardial systole and diastole and inhibit the myocardial remodeling,which may be related to down-regulating the protein expression of TNF-α.
