摘 要:目的:研究生长激素(GH)对脓毒症大鼠肠道细菌易位的影响。方法:将72只大鼠随机分为对照组、GH组、脓毒症组和脓毒症+GH组,再将各组随机分为空肠组、回肠组和结肠组。空肠组大鼠取Treitz韧带处空肠置管,距离Treitz韧带25 cm处造口;回肠组大鼠自回盲部向上25 cm处置管,末端回肠造口;结肠组大鼠行盲肠置管。脓毒症组采用腹腔注射内毒素(LPS)5 mg/kg建立模型,GH组和脓毒症+GH组皮内注射GH1 U/kg。各组分别于肠道置管造口处灌入绿色荧光标记的大肠杆菌。24 h后,在荧光显微镜下观察大鼠肠系膜淋巴结中标记细菌的数量,用流式细胞仪测定肠黏膜上皮细胞的凋亡率。结果:脓毒症时,大鼠肠系膜淋巴结中荧光细菌计数和肠黏膜上皮细胞凋亡率增加,回肠组更加明显。GH可减少脓毒症大鼠细菌易位,抑制肠黏膜上皮细胞凋亡。结论:GH能降低脓毒症大鼠肠黏膜上皮细胞的凋亡率,减少细菌易位的发生。Objective: To confirm the influence of growth hormone on intestinal bacteria translocation in rats with sepsis. Methods: Seventy-two male Sprague-Dawley(SD)rats,weighing 200-250g,were randomly divided into four groups: control group(n=18),growth hormone group(n=18),sepsis group(n=18)and sepsis plus growth hormone group(n=18).Each group were further randomly divided into jejunum,ileum and colon groups.Murine septic model was established through intraperitoneal injection of LPS,and sepsis plus gowth hormone group was followed by growth hormone.E.coli labeled with green fluorescent protein(GFP) was injected into each intestinal segment through the tube.The mesenteric lymph nodes(MLNs)were taken for frozen section and examined for translocation of GFP labeled E.coli.The apoptosis of intestinal epithelial cells was determined.Results: The bacteria labeled with green fluorescent protein and apoptosis in sepsis group were higher than that in control group,and was highest in the ileum group.In sepsis plus growth hormone group,the bacteria and apoptosis were lower compared with sepsis group. Conclusion: Growth hormone can reduce the apoptosis of intestinal epithelial cells and the incidence of intestinal bacteria translocation in rats with sepsis.
