摘 要:目的探讨α-硫辛酸(α-LA)对糖尿病大鼠肾脏聚ADP-核糖多聚酶(PARP)表达的影响。方法 30只SD大鼠随机均分为链脲佐菌素(STZ)诱导的糖尿病组(A组)、α-LA干预的糖尿病组(B组)及正常对照组(C组)。8周后测各组大鼠肾脏超氧化物歧化酶(SOD)、丙二醛(MDA)含量,TUNEL法检测肾脏细胞凋亡,免疫组化法观察PARP在肾脏中的表达,Western blot法检测肾脏半胱氨酸蛋白酶3(Caspase-3)的表达。结果与C组比,A组大鼠肾脏SOD活性下降,MDA含量、肾脏细胞凋亡数、PARP和Caspase-3的表达均显著增多(P<0.05),而B组上述指标均较A组明显改善(P<0.05)。结论α-LA对糖尿病大鼠肾脏保护作用可能与抑制氧化应激、下调PARP表达而对抗细胞凋亡有关。Objective To investigate the effect of α-lipoic acid(α-LA) on the expression of poly-ADP-ribose polymerase(PARP) in the kidney of diabetic rats.Methods Thirty SD rats were randomly and equally divided into three groups of A[streptozotocin(STZ)-induced diabetes mellitus(DM) model],B(DM treated with α-LA) and C(normal control).After 8 weeks,superoxide dismutase(SOD) and malondialdehyde(MDA) were detected in the kidney.The cell apoptosis in glomerular and tubular was counted by TUNEL immunofluorescence assay.The expressions of PARP and Caspase-3 were detected by immunohistochemistry and Western blot,respectively.Results Compared with group C,the activity of SOD was decreased,the content of MDA,the number of cell apoptosis and the expressions of PARP and Caspase-3 were all significantly increased in group A(P0.05),which were much more improved in group B than those in group A(P0.05).Conclusion α-LA protects the kidney of DM rats probably by reducing cell apoptosis via inhibiting oxidative stress and decreasing the expression of PARP.
