Matrigel凝胶和壳聚糖/磷酸甘油凝胶两种支架体外构建组织工程软骨的比较

摘　　要：目的探讨适合构建组织工程软骨的细胞支架材料。方法将体外分离培养的兔关节软骨细胞种植于Matrigel凝胶和壳聚糖/磷酸甘油(C/GP)凝胶两种支架上体外培养3周,对组织工程软骨进行大体标本、HE染色,ELISA检测培养上清液中透明质酸和Ⅱ型胶原的含量。结果软骨细胞在两种支架内均生长良好,Matrigel凝胶中软骨细胞培养3周后仍能维持软骨细胞的表型,其分泌透明质酸和Ⅱ型胶原的能力较C/GP凝胶中软骨细胞强,并可更好的促进受损的软骨细胞增殖。结论 Matrigel凝胶和C/GP凝胶均可作为软骨细胞载体修复关节软骨缺损。Matrigel凝胶支架更适合软骨细胞的贴附生长,有利于维持软骨细胞的表型,是较好的软骨组织工程细胞支架。Objective To compare the effectiveness and feasibility of allograft chondrocytes embedded in matrigel gel and chitosan/glycerophosphate gel（C/GP） scaffolds for articular cartilage repair in rabbits.Methods The complexes of seeding of chondrocytes encapsulated in collagen gel into matrigel gel and C/GP scaffolds were cultured for 3 weeks in vitro.Some complexes were observed with phase contrast microscope and others were allotransplanted into articular cartilage defects in rabbits.The specimens harvested on the 4th week were analyzed by observation,histology and type Ⅱ collagen immunohistochemisrty.Results Chondrocytes were embedded in extracellular martrices largely and evenly when cultured for 3 weeks in vitro.Cartilage-like tissues formed in the articular defects were smooth and well-integrated with adjacent tissues in matrigel gel and C/GP.The results of ELISA examination were strongly positive with type Ⅱ collagen antigen expression in matrigel glue group more than that in the C/GP group.Matrigel gel bracket fitted for the demand of the cartilage reparation in tissue engineering.Conclusion Matrigel gel and C/GP scaffolds can serve as the effective vehicles to delivery chondrocytes to atrticular cartilage defects.Compared to C/GP scaffolds,matrigel gel is better as a carrier of tissue engineered translation acrtilage.
