摘 要:目的观察泡球蚴(Em)感染对Balb/c小鼠肝脏转化生长因子β1(TGF-β1)及其受体TβRⅠ/Ⅱ和信号蛋白Smad2/3表达的影响。方法 Balb/c小鼠随机分为实验组和对照组,泡状棘球蚴原头蚴混悬液肉眼直视注射肝脏诱导小鼠肝泡球蚴感染模型,对照组以相同方法注射生理盐水。于感染第60天处死小鼠,取肝脏组织。用RT-PCR检测肝组织TGF-β1、Smad2、Smad3 mRNA表达;Western blot和免疫组织化学技术检测肝组织TGF-β1、TβRⅠ(TGFβⅠ型受体)、TβRⅡ(TGFβⅡ型受体)、Smad2/3在肝内的表达及定位;HE和Masson染色,光镜下观察肝组织病理改变和肝组织纤维化程度。结果与对照组小鼠比较,Em感染小鼠肝组织内TGF-β1、Smad2、Smad3 mRNA表达升高。Em感染后TGF-β1、Smad2/3蛋白表达增加。TGF-β1、TβRⅠ与Smad2/3阳性反应主要位于肝细胞浆,TβRⅡ主要在肝细胞膜表达,上述指标在两组间表达具有显著性差异。结论在Em感染的肝组织中TGF-β1、TβR及Smad2/3蛋白表达均增高。TGF-β1/Smads信号蛋白参与了Em感染致肝纤维化及肝损伤过程。Em感染可能通过TGF-β1/Smads通路介导宿主与寄生虫间的共生。This study aimed to evaluate the effects of Echinococcus multilocularis(Em) infection on the expressions of TGF-β 1,TGF-β receptors(TβRⅠ and TβRⅡ) and signal proteins(Smad2 and Smad3) in the liver of Balb/c mice.We randomly divided Balb/c mice into an experiment group and a normal control group.The infection model was induced by injecting the Em suspension directly,while the control group was injected with physiological saline.The mice were euthanized and the livers were collected at 60 days.The mRNA of TGF-β1,Smad2,and Smad3 were analyzed by RT-PCR;the protein expression and location of TGF-β1,TβRⅠ,TβRⅡ,and Smad2/3 were analyzed by Western-blot and immunohistochemistry respectively.The level of liver fibrosis and pathological alteration were observed by HE and Masson staining.We found the mRNA and protein expression of TGF-β1 and Smad2,and Smad3 were increased in experiment group,compared to controls.TGF-β1,TβRⅠand Smad2/3 mainly situated in the cytoplasm,while TβRⅡ located in the cell membrane.The expressions of all indexes stated above were significant different between experiment group and control group.All the results showed that the pathway of TGF-β1/Smads signal protein participate the liver fibrosis and hepatic injury in Em-infected Balb/c mice,and the mutualism of host and parasite possibly mediated by the TGF-β1/Smads signal pathway.
